| Objective:1.To establish an experimental model for Helicobacter pylori infection by intragastrically challenging conventional Mongolian Gerbil model after antibiotic pretreatment;2.To evaluate the therapic efficacy of Bacillus acidi lactici to Helicobacter pylori-related gastritis in clean grade Mongolian Gerbil model, through comparing to the efficacy of available trigeminy therapy;3.To explore therapic mechanism of Bacillus acidi lactici to Helicobacter pylori-related gastritis by studying the expression of protein level of mice gastric mucosa COX-2,IL-6,TNF-a..Methods: A total of 32 clean grade Mongolian Gerbil model were randomly divided into 5 groups.The first to the fourth group of mice were infected with Helicobacter pylori to establish the gastritis model and the fifth group was normal.The first group was treated with available trigeminy therapy,The second group was treated with Bacillus acidi lactici strain,The third group was treated with Bacillus acidi lactici and PPI,and the fourth group of mice was treated with sodium chloride solution.the mice were sacrificed 4 weeks post-treatment.The infection of Hp in gastric mucosa of the mice was analyzed by rapid urease test , Giemsa staining and culture. The histopathological changes in the gastric mucosa of mice were assessed in hematoxylin-eosin stained sections. then the expression of protein level of mice gastric mucosa COX-2,IL-6,TNF-a were examined by immunocytochemical staining assay .Results:1.Mongolian Gerbil model with Hp infectious gastritis induced was successfully set up, At 4w, 8w Hp colonization rate was 100% in the experimental mice.2. The eradication of Helicobacter pylori of The first and the second group was 82.5% and 75%respectively,the third group was 75%.There is no significant difference between the first and the second group and the third group in eradication of Helicobacter pylori.3. The chronic inflammatory response in stomach of the fouth group was more serious than the first and second and third group(P<0.05).There is no significant difference between the first and second and third group in the chronic inflammatory response (P>0.05) .4. In protein level, there was no IL-6 in the normal group, COX-2,IL-6,TNF-a in stomach of the first and the second and the third group were significantly decreased comparise to the fourth group(P<0.05). but were significantly increased comparise to the normal group(P<0.05).Conclusion:1.The gastritis model in clean grade Mongolian Gerbil model challenged with Hp SS1 was successfully established in our study. 2.The dysbacteria caused by antibiotics helped colonization of Hp and occurrence of gastritis, The therapy of Bacillus acidi lactici to Helicobacter pylori-related gastritis in clean grade Mongolian Gerbil model was effective.There is no significant difference between Bacillus acidi lactici and available trigeminy therapy.3.0ur results suggested that The treatment mechanism of Bacillus acidi lactici to Helicobacter pylori-related gastritis was concerned with decreasing the expression of C0X-2,IL-6,TNF-a, The study also indicated that Bacillus acidi lactici might be a potential medical component for inhibiting Hp infectious gastritis. |
