[Objective]To investigate the effects and mechanism of nolaterex combined with thalidomide on the growth of gastric cancer in nude mice implanted with gastric cancer cell SGC-7901[Methods]In this experiment the 20 Balb/c nu/nu mice,which had been inoculated human gastric adenocarcinoma cell line subcutaneously,were divided into four groups in random and received treatment subcutaneously every other day,Group A (nolaterex 120mg/kg),group B(thalidomide 100mg/kg),group,C(TNP-470 Nolaterex 120mg/kg+distoval 100mg/kg)and group,D(5%GS).After 3 weeks,the mice were killed and the tumors were immediately removed for further studies.The weight of tumors were calculated,and the Fas,COX-2 and VEGF expression on tumors were detected by immunohistochemisty.Microvessel density(MVD)was counted by endothelial cells immunostained by anti-CD34 antibody.The TUNEL assay was used to examine the apoptosis of tumor cell in nude mice after treatment with nolaterex,thalidomide,both nolaterex and thalidomide group and 5%GS.[Results](1)In nolaterex.,thalidomide,combination group and control group,the data showed that weights of tumor were 2.890±0.143g,3.895±0.221g,2.069±0.260g and 4.227±0.247g respectively;The nolaterex-treated tumors were lower in weight than those in control group(P<0.01).(2)Compared with the control and thalidomide group, the protein of Fas expressed more higher than that in nolaterex and combination group tumor cells(P<0.01).Compared with the control and nolaterex group,the protein of VEGF and COX-2 expressed were lower than that in thalidomide and combination group tumor cells(P<0.05).The amount of MVD in the thalidomide and combination group tissues was significantly lower than that in nolaterex and control group(P<0.01).The apoptosis index(AI)of tumor cells with nolaterex-treated group were higher than that with 5%GS-treated group(P<0.01).[Conclusion]nolaterex can inhibit the growth of gastric cancer.The mechanism of the implanted tumor growth may be associated with induction cell apoptosis. thalidomide inhibits the angiogenesis of tumor and the combinating therapy of nolaterex with thalidomide shows more marked inhibition on the growth of gastric cancer.
