Objective: Bronchial asthma, briefly called asthma, is chronic airway inflammatory in which many kinds of cells such as eosinophils, mast cell and airway epithelial cells, and cellular components. Asthma is a kind of trachea-bronchial tube excessive reactivity disease which causes by the antigenicity or the non-antigenicity stimulation. Modern medicine has many methods to control the symptoms, but the medicine side effect is big and the recurring easy. Traditional Chinese medicine in treating asthma has its own advantages. Therefore it is significant to use cooperation method of Chinese and Western medicine to treat asthma and to research and develop effective medicine.After many years'clinical researches, my tutor found that asthma of children had the pathogenesis feature of the actual situation mixture, therefore she drew up has ventilated and smoothed the lungs the phlegm reduction, solidly this to breathe heavily evenly governs the law. So she herself set a new prescription-Lingma-gubenpingchuan devotion, simultaneously coordinating western medicine to treat asthma, and the curative effect is remarkable. For inquirying into the efficacy and the mechanism of function of the prescription, we proceded the animal experiments. This experiment uses the sensitization law to duplicate the asthma rats model. After observing the influence of the Lingma-gubenpingchuan devotion to asthma rats in the T lymphocyte subgroup, the immunoglobulin and the complement, the tumor necrosis factor as well as the lung organization pathological section influence in order to provide theoretical foundation for the clinic application of asthma.Method: The making model process followed the method used by Cui Longping. 50 healthy Sprague - Dawley rats were randomly divided into five groups: normal control group(normal group), asthmatic model group (model group), Lingma-guben- pingchuan devotion group(Traditional Chinese medicine group, LM group), Aminophylline group (Modern medicine group), drug combination group(union group), each group of 10 rats. In addition to the normal control group, in the first, the fifth and the eighth day, the other four group were injected at abdominal cavities with the antigen fluid 1ml. Meanwhile interjecents cause the rats being allergized. The normal control group were treated with parts equal normal sodium instead of antigen liquid. In the fourteenth day, rats were placed in the plexiglass box incompletely closed, stimulated by 1% egg albumin with ultrasonic atomizer. The normal control group was treated with parts equal normal sodium instead of OVA. The seventeenth day, in addition to the normal control group and model group, the other three groups were administrated before being stimulated prior to half an hour. LM group were filled stomachs by LM. Aminophylline group were filled by Aminophylline suspension. Drug combination group were filled by LM and Aminophylline suspension. The normal control group were filled with parts equal normal sodium one time a day for 8 days. The twenty-fifth day, in 12 hour after the last stimulating, each group were got blood after behead death to detect T-lymphocyte CD4+, the CD8+ percentage, the CD4+/ CD8+ ratio with flow cytometry, detect the immune globulin IgA, IgG, IgM and and the complement C3, C4 with Immu- noturbidimetry, detect the cytokine of TNF-αwith ELISA. After behead death, takes out the left lung immediately, then take the lung apex part the approximately, embed slices, make conven- tional HE dyeing manufacture pathological section, observe the morphological change of lung organizes of each group under the light microscope.Results:1 The effects of LM on pathology morphology in lung organization in asthmatic ratsNormal group: The structures of bronchial tube and lung organization are normal.Model group: There are many EOS and lymphocytes, around the small bronchial tube's pipe wall and the partner artery. The increasing of the bronchial tube mucous membrane rugosofusalina extense, and the proliferation of cup-shaped cells is obvious. LM group and Aminophylline group: The infiltration of inflammatory cells is light or moderate, and we can still see some acidophilia granular cells and lymphocytes. The damage of bronchial tube mucous epithelium is moderate, and the proliferation of cup-shaped cells is not obvious.Union group: The inflammation changes better obviously, and the infiltration of inflammatory cells around bronchial tube and perivascular is light, and we can seldom see the acidophilia granular cells. The mucous epithelium of bronchial tube is complete basicly.2 The effects of LM on peripheral blood T-lymphocyte in asthmatic ratsCompared with the normal group, model group increased in CD4+(P<0.01), and decreased in CD8+(P<0.01), and increa- sed in CD4+/CD8+(P<0.01). Compared with the model group, every treatment groups decreased in CD4+ (P<0.01 or P<0.05), and increased in CD8+(P<0.01 or P<0.05), and decreased in CD4+/CD8+(P<0.01 or P<0.05). Compared with LM group and Aminophylline group, drug combination group decreased in CD4+(P<0.05), and increased in CD8+(P<0.05), and decreased in CD4+/CD8+(P<0.05).3 The effects of LM on the immune globulin IgA, IgG, IgM and and the complement C3, C4 in serum in asthmatic ratsCompared with the normal group, model group increased in IgG (P<0.01), and decreased in IgA,C3,C4(P<0.01), and IgM did not change obviously(P>0.05).Compared with the model group, every treatment groups decreased in IgG(P<0.01 or P<0.05),and increased in IgA,C3,C4(P<0.01 or P<0.05), and IgM did not change obviously(P>0.05). Compared with LM group and Aminophylline group, drug combination group decreased in IgG(P<0.05), and increased in increased in IgA, C3, C4(P<0.05).4 The effects of LM on TNF-αin serum in asthmatic ratsCompared with the normal group, model group increased in TNF-α(P<0.01). Compared with the model group, every treatment groups decreased in TNF-α(P<0.01 or P<0.05). Com- pared with LM group and Aminophylline group, drug combina- tion group decreased in TNF-α(P<0.05).Conclusion:1 LM can adjust the CD4+/CD8+ ratio effectively, reduces IgG, TNF-αthe level, and elevates IgA, the complement level. Thus it plays the anti-gas channel inflammation and the immu- nity adjustment role.2 LM can improve the damage of lung organizations in asthmatic rats obviously.
