Objective:In the states of critical trauma,infection,opera- tion,radiotherapy,chemotherapy and so on,the structure and the function of intestinal trace is changed which causes the damage of intestinal mucosal barricade,and the translocation of the bac- teria and endoxin.The gastrointestinal tract is not noly critical organ that easily damaged,meanwhile,it educes important effe- ct in the development of MODS.And called power organ of MODS or one of spigots that stimulate systematic Inflammato- ry reaction.The increase of mucosa permeability owing to dire- ct or indirect damage is main cause that produce the translocat- ion of the bacteria and endoxin.Therefore,how to effectly prot- ect the gastrointestinal mucosal barrier in various stress is one of the aims of the scholars all over the world to study.The chemotherapy is important ingredient in systematical therapy of malignant tumor.But its adverse effects is not rare to see.among myelosuppression and reaction of Gastrointestinal mucosa are most common.In the process of Chem,due to the reaction of Gastrointestinal mucous membrane from chemothe- rapyeutics,the patients may show up nausea,emesia, diarrhea, severely membrana mucosa may be cellular necrosis,ablate and the patients will show up hemafecia pantosomatous adverse effect.That will break off chems to influence clinical curative effect.How to prevent beforehand and relieve adverse effect is already important topic in curing the patients.Rhubarb is the commonly used Chinese native medicine. The traditional medicine mainly use its swift activeness, to pro- tect and enhance intestinal mucosal barricade,prevent the transposition of the bacterium and the endotoxin,in terms of theory,it reduces the damage of chemotherapy.Recently the res- earch indicate that the Chinese rhubarb has the good protective function to the stomach and the intestines mucous membrane and has the obvious inhibitory action to each kind of tumor cell.But,it is rarely reported that using rhubarb in chemotherapy period.This experiment the Arab League mildew element che- motherapy period blood endotoxin,the bacterium transposition. So we design the test to study the principle of how the rhubarb affect the gastrointestinal mucosa and find the ways to prevent the intestisal mucosa from damaging in the period of chemoth- raphy.Methods: Sixty Wistar rats were randomly and averagely divided into three groups: A:control group, B:ADM group, C:ADM+rhubarb group.After the rats were weighted,the rats in group C were given rhubarb(100mg/kg),group A and B the same doses of saline solution intragastric administrate respect- tively from day 1 to day 6.Group B,and C were injected AD- M(8mg/kg) introperitoneally on the fourth day,respectively to make the model of gut barrier injury.At the same time,group A received the same doses of saline solution injection introperito- neally.The content of endotoxin in peripheral blood was meas- ured in all groups after rats were weighted and killed on the seventh day.The mesenteric lymph nodes(MLN) and livers were harvested for comparing bacterial translocation incidence among four groups.Ileum and colon tissues were taken for pat- hologic examination including comparing mucosal thickness, illus height and width and morphology change of ileum and the weight of each group before and after the test.Statistical Analysis System 6.12(SAS6.12) was used in the study.Analysis of variance and SNK-q test were used for quan- titative data analysis,and Fisher's least-significant-difference test for qualitative data.P values that were less than 0.05 were considered statistical significance.Results:①Compared the weight of each group before and after the test:There was not significant difference among group A (346.20±12.72g),group B(345.45±12.97g),groupC (350.4±12.42g) before test(P>0.05).After test,there was not si- gnificant difference between group A(347.65±12.79g)and grou- p C(345.6±13.17g)(P>0.05).However, the value in group A and group C was significant more than group B(333.90±13.85g )(P <0.05).②The content of endotoxin in peripheral blood of each group:No significant difference is found between group A(0.037±0.011 Eu/ml)and group C(0.041±0.013 Eu/ml (P> 0.05).The value of group A and group C was significantly low- er than that of group B(0.074±0.012 Eu/ml )(P<0.05).③The value of ileum villus height in each group:It was significantly higher in group A(365.22±16.50μm) than that in group B(289.6 8±13.71μm) and group C (334.75±12.37μm)(P<0.05).That of group C were significantly higher than that of group B(P<0.05) .④The value of ileum villus width in each group:It was signi- ficantly wider in group A(174.81±18.19μm) than group B(12 2.70±10.86μm) and group C (156.21±14.40μm)(P<0.05).It was significantly wider in group C than group B(P<0.05).⑤The value of ileum mucosal thickness in each group:It was signi- ficantly thicker in group A(541.23±28.45μm) than group B (422.88±17.84μm) and group C(492.73±23.32μm)(P<0.05). That of group C was significantly thicker than that of group B (P<0.05).⑥The value of colon mucosal thickness in each group:It was significantly thicker in group A (420.10±31.77μm) than group B(264.83±29.24μm) and group C(355.17±43.99μm) (P<0.05).That of group C was significantly thicker than that of group B(P<0.05).⑦T he positive cases of bacterial translocat- ion of MLN were found in group A(0/20),group B(12/20),group C(0/20) and the liver tissue were found in group A(0/20),group B(14/20),group C(2/20).No significant difference was found between group A and group C(P>0.05). The rate of bacterial translocation in group A and group C was significantly lower than that in group B (P<0.05).Conclusions: 1.The rat's gastrointestinal mocosal is dam- aged during the introperitoneal themothraphy of ADM.2. Rhu- barb can protect rat's gastrointestinal mocosal against the dam- age caused by the introperitoneal themothraphy of ADM. 3. How can increase the curative effect and the reduced side effect will be next direction of this domainal research.
