Objective: The establishment of the abdominal aortic aneurysm model is necessary for studying pathogenesis and treatment of the abdominal aortic aneurysm. Yet there is no relative report of the establishment of the abdominal aortic aneurysm model in rabbit in China. This experimental study is to establish an abdominal aortic aneurysm model in rabbit for researching the pathogenesis and treatment of the abdominal aortic aneurysm. Methods: 20 healthy male rabbits were randomly divided into 4 groups. Tow experimental groups were underwent perfusion of the abdominal aorta with porcine pancreatic elastase, the other two contrast groups were underwent perfusion with saline solution. On postprocedural day 2,7 respectively, the surviving rabbits were underwent laparotomy, the diameter of the aorta was measured, and the aortic tissue was excised for histologic examination. Results: The diameter of the porcine pancreatic elastase infused aortas progressed to aneurysm dimension on postprocedural day 7. The creation rate of abdominal aortic aneurysm models was 100% and the mortality rate was 20% in this experimental study. Microscopically, there was total or subtotal loss of elastic tissue and marked inflammatory cell infiltration of the aortic wall in the porcine pancreatic elastase-infused aortas on postprocedural day 7.Conclusion: The method of establishment of the abdominal aortic aneurysm model in rabbit mentioned above is simple and effective. This model exhibits several histologic similarities with human abdominal aortic aneurysm. |
