ObjectiveTo investigate the effect of arcuate nucleus(ARC)and opiod peptides on intraocular pressure(IOP)and the mechanism of this effect.Methods:50 healthy and pure NewZealand White rabbits(100eyes)were divided into 4 groups randomly:Ⅰ.Control group(n=4);Ⅱ.Electrical stimulation group(n=17);Ⅲ.DAGO group(D-Ala2,NMe-Phe4, Gly-olenkephalin)(n=15);Ⅳ.DPDPE group([D-Pen 2, D-PenS]-enkephalin)(n=14).Observe the IOP after electrical stimulation in rabbits' arcuate nucleus(ARC)and micro-administration of selective opioid receptor agonists and non-selective opioid receptor antagonist naloxone in ARC under stereotaxic apparatusResults:1.Electrical stimulation in ARC caused obvious decrease of IOP (P<0.05),which could be partly turned over by pretrement of naloxone both in ARC and under conjunctiva 2.Micro-administration of delta and mu opioid receptor agonists caused dose-related decrease of IOP (P<0.05),which could be partly turned over by pretrement of naloxone under conjunctiva 3.Unilaterally electrical stimulation and administration of selective opioid receptor agonist in ARC caused bilateral reduction in IOP,however there was a more significant response in Ipsilateral eye than in contralateral eye.Conclusions:Arcuate nucleus(ARC)takes a part in the negative regulation of IOP in which opioid neuron may be involved.
