ObjectiveBy immunohistochemical staining, Reverse Transcription-PCR and Real-time Fluorescence quantitative reverse transcriptase polymerase chain reaction, we detect the Mucin gene (MUC2,MUC5AC,MUC5B,MUC18 and MUC19)expression in the nasal polyps, allergic rhinitis (AR)and the normal nasal mucosa of patients. Analyse the relationship of Mucin gene in nasal polyps and Allergic Rhinitis patients. Discuss Mucin gene in the pathogenesis of nasal polyps and Allergic Rhinitis patients and the role and clinical significance.MethodsAcquisition of 35 cases of nasal polyps, 25 cases of allergic rhinitis inferior turbinate and 20 cases of simple nasal septum deviation of normal epithelial inferior turbinate. Specimens were divided into two groups, one group was fixed by 4% POM, conventional dehydration, embedded in paraffin, slicing a row, then staining by Elivision method to analysis the MUC expression in nasal polyps and allergic rhinitis patients, and the other group was used to extraction of total RNA, reverse transcription for cDNA, a housekeeper geneβ-actin gene as the internal standard for RT-PCR(reverse transcription-polymerase chain reaction) and real-time quantitative PCR reaction, detected in the samples MUCS gene expression level in a separate specimens, another was extracted organizations total protein, Western hybridization blotting, housekeeper geneβ-actin gene as the internal standard, Determination with the purpose of the optical density (OD), andβ-actin optical density (OD) ratio for representative of MUCS the relative protein expression levels.Results1. By immunohistochemical staining observed in the nasal tissue distribution MUC2, MUC5AC, MUC5B, MUC18, showed the 4 MUCS in nasal polyps, allergic rhinitis patients and normal mucosa in all the expression of positive particles were mainly concentrated in the nasal epithelial cells in the cytoplasm. MUC5AC, MUC5B in nasal polyps and AR patients expression was significantly higher than that in normal mucosa (P<0.05). The expression of MUC5AC, MUC5B in nasal polyps was no significant difference with that in AR patients(P>0.05). The expression of MUC2,MUC18 in nasal polyps and AR was no significant difference with that in normal mucosa (P>0.05).2. We used RT-PCR detected the gene expression of MUCS in the nasal polyps tissue and AR mucosa then analyze the difference of normal mucosa. MUC5AC,MUC5B in nasal polyps and AR patients expression was higher than that in normal mucosa (P<0.05). The expression of MUC5AC,MUC5B in nasal polyps was no significant difference with that in AR patients(P>0.05). MUC19 in AR patients expression was higher than that in nasal polyps and normal mucosa (P <0.05), the expression of MUC19 in nasal polyps was no significant difference with that in normal mucosa (P>0.05).The expression of MUC2, MUC18 was no significant difference within the three groups (P>0.05).3. Established a successful real-time quantitative RT-PCR detection methods, amplification efficiency (>95%), reproducibility and stability, MUC5AC,MUC5B in nasal polyps and AR patients expression was significantly higher than that in normal mucosa (P<0.05). The expression of MUC5AC,MUC5B in nasal polyps was no significant difference with that in AR patients(P>0.05). The expression of MUC2, MUC18 in nasal polyps and AR was no significant difference with that in normal mucosa (P>0.05).4. The expression of MUC2,MUC5AC,MUC5B,MUC18 was different in nasal polyps group, AR group and in normal group(P<0.001). The results were: the tendency of mucins secretion was mainly homological in nasal polyp,allergic rhinitis and normal group. Especially among the total groups,the secretion of MUC5AC was the maximum, and MUC5B was the second.Conclusions1. All the MUC2, MUC5AC, MUC5B, MUC18, MUC19 express on nasal mucous, and the expression of MUC5AC and MUC5B were higher than the other three.2. There was no difference of the expression of MUC2 and MUC18 in nasal polyps group, AR group and in normal group.3.The results of immunohistochemistry, RT-PCR and FQ-RT-PCR showed that the expression of MUC5AC,MUC5B in nasal polyps group and AR group were higher than normal group. The secretion of MUC5AC,MUC5B in nasal polyps group and AR group was on high level. The expression of MUC19 in allergic rhinitis was higher than that in nasal polyps group and normal group. It indicated that the secretion of MUC19 in allergic was on high level.
