Both the increased threshold of brainstem evoked potential and decreased amplitude and derivation rate of distortion product otoacoustic emission are observed in ID animals. It has been found that Tiebifu infusion have the best curative effects in deficient kidney-induced hearing loss over the past 20 years. Protein is a fundamental component involved in all aspects of cellular processes, as well as aspects of human health and diseases. We can explore the mechanism of the organ and tissue by identifying the architecture and the function of the protein.Objective(1) To study the effects of Tiebifu infusion on the protein expression of the cochleas in rats with SSHL of ID-induced Shenxu.(2) Explore the molecular mechanism of kidney-tonifying method in the treatment of deficient kidney-induced hearing loss.(3) Find further evidences of the biochemical basis for the relationship between kidney and ear.MethodsAccording to the experiments have ever done, it is know that there have been some change in the protein profile of the cochleas in rats after the kidney-tonifying method been used. So the Tiebifu infusion is used to treat the iron deficiency rats. Then the tecknowledge of proteomics is used to test the protein expression of the cochlea to find the difference. The yonger Wistar rats that weighted 25 to 35g, have no diseases were selected. Put them into 3 groups at random. The normal group were fed with normal dietary for 16 weeks. Others were fed with iron deficiency diet for 8 weeks to make iron deficiency hearing loss models that ABR threshold raising≥15dB at least in one ear . Then they were randomized into 2 groups that called iron deficiency group and Tiebifu group, the formers rats were fed with iron deficiency diet for another 8 weeks, the laters were fed with Tiebifu diet 8 weeks. Then we test the protein of cochlea to find the differences and search for data file using BIOWORKS softwares.Results873 proteins were detected in cochleas of Tiebifu group. 989 and 1019 proteins were detected in cochleas of iron deficiency group and normal group respectively. Every group also detected many proteins of their own. Each group of protein were classified in many degrees according to cellular component, biological process and molecular function. Actin, aortic smooth muscle; Actin, cytoplasmic 1; Isoform 1 of Titin turn up after Tiebifu was used. Cochlin Precursor protein was found in Tiebifu group, its'Pep Count Sequence is 629, and its'Cover Percent is 40.07%. Other proteins' PepCount Sequence are below 50, the CoverPercent are below 30%. 46 kDa protein,48 kDa protein and other new protein are found in Tiebifu group, their functions are unknown, and they also are not named, but their Cover Percent are higher.Conclusion1. 46 kDa protein,48 kDa protein and other new proteins turn up in the rats after Tiebifu medication.2. The effective role of Tiebifu infusion is proved to be linked with many proteins like actin, myosin, titin and cochlin precursor in cochlea.3.
