| Squamous cell carcinoma is one of Head and Neck Neoplasms. Three percent of human cancer is laryngeal squamous cell carcinoma, the amount of this kind of cancer is increasing these years. It is very significant to study the generation, metastasis and recurrence of laryngeal squamous cell carcinoma.Galectin-3 is a member of a growing family ofβ-galactosidebinding animal lectins. This protein is expressed in a variety of normal and tumor cells. It plays a crucial role in tumor cells adhesion, proliferation and differentiation. Recent studies indicate expression of galectin-3 is related with tumor transformation, but the mechanism is not clear.The expression of galectin-3 and VEGF were detected by using immunohistochemical in twenty-nine cases of LSCC and eighteen cases of laryngeal benign lesion tissues.The expression of galectin-3 is remarkably higher in LSCC than in normal and benign lesion tissue, and there is statistical significance (P<0.05) between LSCC and normal tissue. The positive expression of galectin-3 is in inflammatory cell in benign lesion tissue, however, in LSCC galectin-3 is in all kinds of cells and the positive degree is higher.The expression of VEGF is higher in LSCC than in normal and benign lesion tissue, and there is statistical significance (P<0.05).Moreover,there was also a statistical positive correlation (r=0.423, P<0.05) between the expression level of galectin-3 and VEGF.We also analyze relationship between galectin-3 and VEGF with clinical stage, histodifferentiation and Neoplasm Metastasis. The results indicate that we found that the tissue differentiation level was in coincidence with the ratio of galectin-3 positive expression (P<0.05). The expression of VEGF is different in clinical stage, histodifferentiation and Neoplasm Metastasis, but there is no statistical significance. Our study analyze the relationship between galectin-3 and VEGF with tumor growth, differentiation, and metastasis. This study played a fundamental role in deeply investigating the effect of galectin-3 and VEGF in tumorigenesis and cell proliferation, and the relationship between them with human cancers. It will supply a new clue to prevent and treat the associated diseases. It possesses significant theoretical meanings and application values to design and develop a new type of micromolecule therapeutic medicine blocking or activating some signal transduction pathways.We expose HEP-2 cell to different concentration of staurosporine for different time. The Hep-2 cell viability is measured by MTT assay. The apoptosis cells were observed by light microscopy and fluorescence microscopy. Expression of caspase-3 is detected by using western blot with anti-casepase-3. We also stimulate hep-2 cells with both resveratrol and staurosporine. Then we found that resveratrol can enhance cell apoptosis induced by staurosporine.This study establish the apoptosis hep-2 cell model with staurosporine. It possesses significant theoretical meanings and application values in studying the mechanism of apoptosis. |
