Objective: It has been demonstrated,neural stem cells in situ in the adult rats could been actived after cerebral ischemia preconditioning,limb ischemic preconditioning (LIP) performed immediately prior to cerebral ischemia could protect pyramidal neurons of the CA1 hippocampus from ND normally induced by cerebral ischemia/reperfusion. further, neural pathway participated in the protective effect of LIP on brain.in the experiment, We investigated the active effects of LIP on the neural stem cells in hippocampus in the adult rats after cerebral infarction, and the role of Neural pathway participated on the mechanism of LIP actived of neural stem cells. Method: We use the models of the rats that right middle cerebral artery were obstruction to observe neural stem cells'quantitative diversity of the hippocampus in rats of cerebral infarction after LIP.LIP was performed by occluding the right femoral arteries for 10 min 3 times in an interval of 10 min.for the purpose, 60 SD rats were assigned to 5 groups by random:①control(n=12), non occluding the right femoral arteries only exposure, and cerebral infarction,②LIP + cerebral infarction,being the interval time between LIP and cerebral infarction, the group was further divided into 12h, 24h and 48h group(n=12),③femoral nerves section(FNS) + LIP + cerebral infarction(n=12). Nestin positive cells'quantitative diversity of hippocampus in rats were examined 24h after cerebral infarction under immunohistochemistry staining.Nestin was used to identify neural stem cells. Random select three field of vision under high power microscope to counter nestin positive cells,the average of number was calculated as numerical data. Result: nestin positive cells in hippocampus of control group: 27.33±3.38, compared with the controls,the number of nestin positive cells after LIP were increased in 12h(P<0.05), significantly increase in 24h,48h.Nestin positive cells in FNS+LIP24h group was no change compared with the control group(P<0.05). It showed that neural stem cells were increased in hippocampus in rats after LIP.But it was no change in FNS + LIP + cerebral infarction group. Conclusion: neural stem cells in hippocampus in rats after cerebral infarction could been actived in 12h-48h after LIP, these celle may be have potential therapeutic effect on self recovery of cerebral infarction. |
