ObjectiveTo observe pregnancy mice's stationary time when forcing swimming test(FST), get to know whether pregnancy mice have more obviously hopeless behavior(stop struggling,sationary time prolongs).At the same time,to know whether the hopeless level of pregnancy mice(length of stationary time)become aggravated after several forced swiming.By the way of half quantitative mensurament(immunohistochemisty,IH)to check out the expressive level of ERαin the pregnancy mice's cerebral cortex after several forced swimming;To simplely analysis the quantitative relation between length of pregnancy mice FST's stationary time and the brain ERα's expression level;And preliminaryly to discuss whether FST's stationary time and cerebral cortex's ERαis straight line correlation.Materials and MethodsSame batch female Kunming mice,source from Chinese Medical University animal experiment center;to divide into conceive group and compare group in radom, and have got 31 pregnancy mices and 27 compare mices;in which,there are 3 pregnancy mices premature labors,do not carry out 6th FST and IH inspection and measurement;Start to carry out FST from 12th day of pregnancy,and record 2th and 6th FST's stationary time.After six times forced swimming,to execute and take brain,have got 28 pregnancy mices and 13 compare mites' brain,to slit with hippocampus as center, make paraffin specimen;to detect ERαof the mice's cerebral cortex by the experiment of immunohisto-chemistry(SP).Statistics analysis adopts SPSS13.0 software;If measure information are not satisfy with normal distridution and variance evenness,inspect with Mann-Whitney U test;Satisfy with overall normal state and homogeneity of variance,adopt two independent sample T-test;Between two variables information,relationship is analysed by the way of Pearson straight liner analysis.ResultsPregnancy mices' stationary time are longer than compare mices' when 2th and 6th FST,Pis 0.011 and 0.016,discrepancy has statistics meaning;Pregnancy mices' stationary time of 6th FST are longer than those of 2th FST obviously,P is 0.016,discrepancy has statistics meaning.In the result of Immunohisto-chemistry(SP)experiment,pregnancy mices have expression of ERαin cerebral cortex as well as compare mices,but its score are higher than compare mices',P is 0.008,discrepancy has statistics meaning.Take pregnancy mices which stationary time>180s when FST as long time group, and those stationary time<180s as short time group,compare 2 groups' score of ERα,P is 0.128,has no statistical significance.In 2th and 6th FST,there is always no linear correlation between stationary time and score of cerebral cortex's ERα,P is 0.800 and 0.607,coefficient correlation have no significance.ConclusionsIn FST,pregnancy mices' stationary time are longer than compare mices', pregnancy mice are prone to depression.After 6 times forced swimming,pregnancy mices' stationary time became long obviously,and can be considered as a model of antepartum depression.The level of ERαin pregnancy mices' cerebral cortex are higher than those of compare mices',its maybe relate to causing of antepartum depression.Have not discovered the quantity-relationship of pregnancy micesbetween FST's stationary time and the score of ERαin IH.And there are no statistical significance between FST's stationary time and the level of ERαin mice's cerebral cortex.
