Hyperlipidemia is the presence of raised or abnormal levels of lipids and lipoproteins in the blood. Lipid and lipoprotein abnormalities are extremely common in the general population, and are regarded as a highly modifiable risk factor for cardiovascular disease due to the influence of cholesterol, one of the most clinically relevant lipid substances, on atherosclerosis. In addition, some forms may predispose to acute pancreatitis.At present, although western medicine of lowing the blood fat has certain curative effect, the side effect is large too, not suitable to take medicine in long-term. So searching and developing the asepsis effective traditional Chinese medicine of lowing the blood fat has important practical meaning.We investigated the effects of polysaccharide from Ganoderma tsugae on hyperlipidemia and atherosclerosis in mouse and rats model, and compared the results of different administration (ig or ip) and different dose groups. Through different administration with oral intubation and intraperitioneal paracentesis and different dose compare with inter groups. Simvastatin was used as positive control. The concentrations of total cholesterol, triglyceride, low density lipoprotein cholesterol and high density lipoprotein cholesterol were investigated by biochemical methods. The effect of polysaccharide on the blood lipid was studied, and the primary mechanism was also discussed.The results showed that the polysaccharides can decrease the levels of TC, TG and LDL-C in serum, and enhance the level of HDL-C.The statistical results showed that the effects of regulated-blood lipid and anti-atherosis are obvious, whether in oral intubation group or in intraperitioneal paracentesis group. Compare with simvastatin group, there was no obvious difference.The expressions of HMG-CoA reductase and FAS mRNA were decreased in different degree by RT-PCR. HMG-CoA reductase is the rate controlling enzyme of the mevalonate pathway, the metabolic pathway that produces cholesterol and other isoprenoids. The expression of ApoAI protein was increased and the expression of ApoB100 protein was immovability by biochemical assay.Conclusion: Polysaccharides can reduce the levels of LDL-C, TC, TG and enhance the level of HDL-C in serum in rats and mouse model of hyperlipidemia, it is effective for regulating blood lipid. Our data indicated the mechanism that Ganoderma tsugae polysaccharides can decrease the expressions of HMG-CoA reductase and FAS mRNA in order to reducing the synthesis of TC and TG to regulate the blood lipid.
