Cholinergic neurons serve a variety of functions including learning,memory and motion processes. Many behavioral and functional messes in organism are involved in disease of cholinergic system. But the investigation on the generational of cholinergic neurons is limited. The relationship between extracellular factors and homeodomain transcription factor that induce cholinergic neurons generation is the key of understanding the mechanisms of it.According to the information , the dorsal ventricular zone (VZ) and the subventricular zone (SVZ)of the telencephalon is a new cortical pyramidal neurons cradle. In the cortex ,the cholinergic neurons is a kind of pyramidal neurons. Choline acetyltransferase(ChAT)is a marker for cholinergic neurons and it can show the generation,transplantation and distribution of cholinergic neurons. The cholinergic precursors'generation is from E12 in mice. Mainly in the ventral side of the elencephalon brain ventricle. It emerge in the dorsal lateral ventricle of the lateral and third ventricles from E14 in mice.. E15 in mice cholinergic neurons also appear in the lateral cerebral ventricle,except above.According to the information of, the radial neuroglia play a dual role in the development of neurons and glial cells , that characteristics of glial cells and the characteristics of neural stem cells ,it not only can be used as the scaffolds of neuronal migration, can be self-renewal , but also generate nerve cells in the early cortex generation by using dissymmetry desintegrate, in the advanced stage development generate astrocytes.It express the signs of nerve cell protein nestin, and it desintegrate in the germinal zone ,the generated neuroblast and moved to the destination along the radial glial cell processes. Vimentin is a marker for radial neuroglia cell and it can show the generation,transplantation and distribution of radial neuroglia cell. There is also information to confirm that, during the neuron development , 96% -98% of the cells in ventricle region were radial neuroglia cell, these reports suggested that radial glial cells may be progenitor cells of the cholinergic neuron.Firstly the telencephalon of rats at embryonic 14 days (E14) were performed coronary sections,then immunohistochemistry double staining for vimentin(a protein marker of radial glial cell) and choline acetyltrans-ferase (ChAT, a protein marker of cholinergic neuron) to test whether ChAT was expressed in the population of radial glial cell in vivo.secondly the telencephalons cell of rats at embryonic 14 days (E14) were primary culture,then Immunocytochemical double staining for vimentinand choline acetyltrans-ferase to test whether ChAT was expressed in the population of radial glial cell in vitro.The E14 rat telencephal brain Vimentin / ChAT double fluorescence immunohistochemical staining showed that: Vimentin-positive cells coexpressed ChAT in ventricular zone of dorsal telencephalon and lateral ganglionic eminence of E14 rat,it is the same as the former information. telencephalons cell of rats at embryonic 14 days (E14) were primary culture,then Immunocytochemical double staining for vimentinand choline acetyltrans-ferase confirm E14 rat telencephal brain cells cultured in vitro with cells Vimentin / ChAT Immunochemistry double staining positive expression.In vivo and in vitro experiments common confirmed that in the E14 rat telencephal brain ChAT was expressed in the population of radial glial cell.The vitro and vivo experiments confirmed that in the E14 rat telencephal brain ChAT was expressed in the population of radial glial cell.The population of radial glial cell indeed has the property of neural stem cell and continually undergoes asymmetrical cell division to generate neuronblast with cholinergic phenotype,or cholinergic progenitor..It is suggested that the cell fate of the population of radial glial cell has been decided in the asymmetrical cell division,and then synthesizes into neuroteansmitter acetyleholine. Providing a theoretical basis for further study cholinergic neurons in the development of mechanisms .
