Objective:To detecte the expression of RhoC and MMP-2 in different epithelial ovarian tissues, and RhoC and MMP-2 in 91 cases epithelial ovarian cancer,we retrospectively analized the relations of RhoC and MMP-2 expression and the age of patients and clinical pathology characteristics.Methods:This text used immunohistochemical method to examine the RhoC and MMP-2 expression situations in 91 cases malignant epithelial ovarian tissues . Take 12 cases of epithelial ovarian benign tumors and 10 normal ovarian tissues as a control, we examined the RhoC and MMP-2 in ovarian cancer in the positioning and qualitative research. Cut specimens, made the immunohistochemical sections, then carried on immunohistochemical staining, read films under the microscope. cytoplasm or the membrane to appear yellow or yellowish brown were believed positive results,which were calculated to obtain the positive rates of two proteins. Two group of positive rate differences were analized withχ2 test , and nonparametric statistics Spearman correlation analysis were used in the correlation analysis of the two proteins in epithelial ovarian cancer tissue.Result:In 10 cases of normal ovarian tissue, 12 cases of benign epithelial ovarian tumor and 91 cases of epithelial ovarian cancer ,RhoC expression rates were 0%, 0%, 37.3% (35/91), and MMP-2 Expression rates were 0%, 0%, 68.1% (62/91). Statistical analysis come to the following conclusions: The expression rates difference of RhoC in epithelial ovarian cancer and benign epithelial ovarian tumors and normal ovarian tissue was significant,. The expression rates difference of MMP-2 in epithelial ovarian cancer and benign expression of epithelial ovarian tumors and normal ovarian tissue also shows a significant difference. 91 cases of patients with epithelial ovarian cancer are in the age distribution between the ages of 18-73, with an average age of 45 years old. Compare RhoC expression between≤45-year-old of epithelial ovarian cancer patients and>45 years old ,χ~2 = 0.18, P>0.05, there was no significant difference . Compare MMP-2 expression between≤45-year-old of epithelial ovarian cancer patients and>45 years old ,χ~2 = 2.54, P>0.05, there was no significant difference. Thus RhoC and MMP-2 protein expression in epithelial ovarian cancer was not related to age.The relationship of two proteins and epithelial ovarian cancer clinic pathological characteristics: RhoC expression compares in different types of pathology organization typeχ~2=1.07, P>0.05, no statistical significance.So there is no significant relationship between the RhoC protein expression and pathological type. RhoC expression in the clinical stagesâ…¢~â…£positive rate (59.1%) was significantly higher than that of stagesâ… ~â…¡(14.2%) (χ~2= 19.62, P <0.005), a comparison of the two, there are significant differences.In the organization differentiation , compared low differentiated of the positive rate (56.6%) with the midst or well-differentiated ovarian epithelial ovarian cancer of the positive rate (13.1%), the difference was statistically significant (χ~2= 17.64, P <0.005); positive rate (62.7%) of transfer cases compared with positive rate (7.5%) of the without transfer cases , the difference was statistically significant (χ~2= 28.9, P<0.005). The expression of MMP2 in different histological types of epithelial ovarian cancer showed no significant difference (χ~2= 3.41, P>0.05);clinical stagesâ…¢~â…£positive rate (95.9%) was significantly higher than that of stagesâ… ~â…¡(35.7%) (χ~2= 37.75, P <0.005).There are significant differences in MMP2 in degrees of differentiation.The MMP2 in the low differentiated (94.3%) was higher than that of midst or well-differentiated (31.5%) (χ~2= 31.78, P<0.005); the positive rate (90.1%) of transfer cases compared with the non-transfer cases positive rate (65%) was higher,the difference was statistically significant (χ~2= 18.83, P<0.005).In 91 cases of epithelial ovarian cancer, 62 cases of RhoC and MMP-2 expression was consistent.In 56 cases of RhoC negative patients and 29 cases of MMP-2-negative patients, 28 cases are the same patients. In 35 cases of RhoC-positive patients and 62 cases of MMP-2 positive patients, 34examples are the identical patients. There is one case with RhoC positive expression and MMP-2 negative expression ; There are 28 cases with RhoC negative expression and MMP-2 positive expression ; the positive expression of RhoC and MMP-2 shows a positive synergy and consistent positive correlation (r = 0.492, P<0.05). The relationship of RhoC and MMP -2 is close, and in epithelial ovarian cancer invasion and metastasis they play an important role. In epithelial ovarian cancer cases, epithelial ovarian cancer showed high RhoC expression, and the expression of MMP-2 was also high,, so there is positive correlation.between RhoC and MMP-2 .91 cases of epithelial ovarian cancer patients were revisited after six months chemotherapy ,and chemotherapy drugsincluded cisplatin and cyclophosphamide, doxorubicin, paclitaxel. 91 cases of epithelial ovarian cancer patients were carried on revisit for three years , the 49 patients were died. RhoC positively expressed in 20 cases, 29 cases is in negative expression.The positive rate was 40.8% (20/49).42 cases of the survival have 15 patients with positive expression of RhoC, 27 negative cases, the positive rate of 35.7% (15/42).compared the two groups, the difference was significant ,χ~2 = 4.27 (P <0.05).during 3 years,in 49 cases of death in patients with MMP-2, positive are 36 cases, 13 cases are negative, the positive rate of 73.4% (36/49).In 42 cases of survival in patients with MMP-2 expression, positive are 26 cases, negative are 16 cases, the positive rate was 61.9% (26/42).the two groups were compared,χ~2 = 27.47 (P <0.005), the difference was significant. RhoC protein and MMP-2protein expression in epithelial ovarian cancer dead patients was higher than that of the survival rate, so the higher the RhoC and MMP-2 protein expressed, the poorer the patients's prognosis was.Conclusion:1. The expression of RhoC protein and MMP-2 protein in normal ovarian epithelial ovarian tissue and benign tumors are negative; in epithelial ovarian tumors are positive, suggesting that ovarian cancer is RhoC protein and MMP-2 Protein expression of the tumor.2. The expression rate of RhoC protein and MMP-2 protein are not related with age, but there are relationship with pathological types, and later clinical stage, poorer histological grade, and have metastasis , The expression of RhoC protein and MMP-2 protein in ovarian epithelial carcinoma patients is higher, suggesting that the expression of RhoC protein and MMP-2 protein in epithelial ovarian cancer is related with the progress and the degree of malignancy, and thus Detection of RhoC protein and MMP-2 protein can be used to determine the extent of malignant ovarian tumor invasion and the ability of metastasis . 3.In epithelial ovarian cancer, RhoC protein and MMP-2 protein expression are relevant, suggesting that they may be joint action in the evolution of epithelial ovarian cancer. The high expression of RhoC protein and MMP-2protein are low rate of survival patients, a poor prognosis. |
