Objective To compare the effects of chitosan and sodium hyaluronate on proliferation of retinal pigment epithelial cells in rabbit, discuss the security of chitosan use as packing in vitreous body cavity and the effect of prevent and cure proliferative vitreo-retinopathy (PVR), prepare for farther research of chitosan use as packing in vitreous body cavity.Method The rabbit RPE cells (passage 2-6) were cultured with various concentrations of chitosan and sodium hyaluronate in DMEM, the final concentrations were 10mg/ml,5mg/ml,2.5mg/ml,1.25mg/ml and 0.625mg/ml. The cells were cultured for 72h, then measured the absorbency by MTT method at the wave of 540nm and determined the rate of inhibition. The cell viability was estimated by trypan blue exclusion test.Result Chitosan at a given concentration (>1.25mg/ml) significantly inhibited the proliferation of cultured rabbits'RPE cells. The inhibition was interrelated with the concentration of chitosan. And sodium hyaluronate at all concentrations hasn't inhibited the proliferation of cells.Conclusion Chitosan inhibited the proliferation of cultured rabbits'RPE cells. It is possible chitosan can be used to prevented the occurring of PVR. |
