The traditional Chinese medicine(TCM), as the precious treasure of the Chinese nation, has made an indelible contribution to the disease prevention and treatment for thousands of years. The traditional quality control methods, such as appearance identification, microscopic identification, physical and chemical identification and assay methods, have played a positive role in the quality assurance of TCM. But the substitutes, counterfeits and adulterants can't be fully distinguished from genuine materials by these traditional methods. Therefore, how to establish a new detection method, which is more effective, faster and can reflect the inherent overall situation of the chemical substances in TCM, has been paied more attention to by scientists. At present the establishment of herbal chemical fingerprint has been internationally recognized as the reliable quality control method of TCM. However, to establish what kind of fingerprints has not gotten a uniform understanding. HPLC fingerprint is a method studied a lot. It can quantitatively detect 7-10 chemical substances simultaneously and reflect more chemical information in TCM. But it can only determinate a certain type of chemical substances in TCM. For some plants from the same genus, with the same main chemical constituents, HPLC fingerprint could not distinguish them sufficiently.Hydrogen nuclear magnetic resonance (~1H-NMR) technology can theoretically detect any hydrogen atom contained in organic components. In TCM, the main effective chemical substances are the secondary metabolites. For the secondary metabolites, establishing a ~1H-NMR method will more fully reflect the inherent quality of natural medicine materials. The quality evaluation of TCM based on ~1H-NMR is still at the exploratory stage abroad and home, which is mainly intuitively spectroscopy or several major signal peaks' comparison. For getting a good ~1H-NMR spectroscopy, there should be a lot of influential factors from equipment, extracting and preparing process, the habitat, collective season and et al. To combine ~1H-NMR with multivariate statistical analysis(MSA), could objectively reflect the inherent quality of TCM.Seabuckthorn, the plants from Hippophae Linn.(Elaeagnaceae), are with great ecological and medicinal values. Up to date, there are only four fingerprint reports about seabuckthorn, three of which are on HPLC fingerprint and one is on capillary electrophoresis fingerprints. In this paper, we try to establish a method based on ~1H-NMR-MSA for the plants of Hippophae Linn.. As the multivariate statistical analysis, principal component analysis (PCA), hierarchical cluster analysis (HCA) and similarity analysis are used.21 berries of seabuckthorn, belonging to three species and five subspecies, have been studied on the difference, clustering and similarity. The results based on the whole spectrum analysis of the 21 samples show good similarities in the samples of H. rhamnoides subsp. sinensis, H. rhamnoides subsp. wolongensis, H. salicifolia and H. rhamnoides subsp. yunnanensis, and in the samples of H. neurocarpa subsp. stellatopilosa and H. neurocarpa subsp. neurocarpa. The results based on ~1H-NMR-PCA show the different between H. neurocarpa subsp. stellatopilosa, H. neurocarpa subsp. neurocarpa and H. rhamnoides subsp. sinensis, H. rhamnoides subsp. wolongensis, H. salicifolia, H. rhamnoides subsp. yunnanensis. The results based on theδ9-5ppm spectrum analysis of the 21 samples show good similarities in the samples of H. rhamnoides subsp. yunnanensis, H. neurocarpa subsp. stellatopilosa and H. neurocarpa subsp. neurocarpa, and show the different in the samples of H. rhamnoides subsp. yunnanensis with H. rhamnoides subsp. sinensis, H. rhamnoides subsp. wolongensis, H. salicifolia. The results of the same subspecies analysis indicate the samples of the same subspecies have good overall similarities. The ~1H-NMR-MSA study of seabuckthorn shows clearly the difference between different subspecies and similarity in the same subspecies. This method is feasible and should have a good prospect in the quality control of TCM.
