Objective To study the basic expression of RARs and CRABPs at the protein level in HaCaT cells and the dose-time effect of AT and Acitretin on them. To study the effect of AT and 1,25-(OH)2D3 on expression of RXRαand VDR at the transcriptive level in HaCaT cells. Methods HaCaT cells were cultured, RARs and CRABPs of each team were detected by western–blot. The mRNA levels of RXRαand VDR of each group were detected by PCR. Results RARα, CRABPⅠand CRABPⅡcould be detected in HaCaT cells, but not for RARβ. The protein levels of RARαand CRABPⅡin HaCaT cells were upregulated by different concentrations of AT or Acitretin, RARβof each group couldn't be detected. CRABPⅠhad no significant dose and time effect with RA. RXRαand VDR could be detected in HaCaT cells, The mRNA level of VDR was markedly upregulated about 8 times by adding only 1,25-(OH)2D3, VDR and RXRαin other groups had no statistical differences compared to the contrast. Conclusion In HaCaT cells, AT and Acitretin could induce RARαand CRABPⅡ, maybe it's related to the RA process pathway. AT may had influence on vitamin D signaling pathway through RXR/VDR heterodimers.
