Study On The Chinese Herb Compound 0326

Chinese herbal compound 0326 is studied by us. Through primarypharmacology test, it was proved to have anti-AIDS activity,improving immunity. The compound is composed of Radix AngelicaeSinensis, Fructus Cnidii,Rhizoma Bletillae,Rhizoma Coptidis, whichhave the effective constituents of ferulaic acid, osthole, berberineand polysaccharides. Main research works and results were reported asfollows:The four crude drugs were extracted respectively, with fixedextraction condition. So the extraction rates were obtained, whichcan solve the content change and unstable pharmacodynamic actioncaused by fluctuation of extraction rate. Radix Angelicae Sinensis,Fructus Cnidii,Rhizoma Coptidis were respectively extracted by 60%alcohol, and then by water. Rhizoma Bletillae was extracted throughpercolation method.The method of HPLC was adopted in the test, the guality controlmethod on the HPLC finger-print of the compound was established. Thecompound was composed of liposoluble constituents and water solubleconstituents. The main water soluble active ingredients werepolysaccharides. We tested the content of polysaccharides by themethod of sulphuric acid-Phenol. The liposoluble constituents weredissolved in 70% methanol, and the quality and content were controlledby the HPLC finger-print method.Pharmacology research indicated that the compound 0326 showed aninhibitory action on HIV-1 reverse transcriptase in vitro. The acutetoxicity of 0326 in vitro was investigated and the value of the IC50 of compound was determined. To observe the effect of 0326, the modelof mouse hepatocarcinoma (H22) solid tumor-bearing mice was built,and treated with the extraction, respectively. The extraction of 0326showed cytotoxic to human adenocarcinoma of lung A₅₄₉ in vitro, and alsoshowed a strong cytotoxic to human cervical carcinoma Hela in vitro.
