| Ilexpubesceus,the dry root of Ilex pubescens Hook.et Am,is commonly used in south China.It is widely used in clinic and has certain effect on cooling the blood, promoting the blood flow and circulation,relieving the pain by detumescence or clearing the heat.Since 1980s,Ilexpubesceus and its compound preparation has been widely used on cardiovascular diseases,especially on anti-throm and removing brain edema induced by cerebral apoplexy during the ischemic cerebral apoplexy.Till today, however,the basic research on Ilexpubesceus was fall behind and lack of perfect method of quality control,therefore the species and quality of Ilexpubesceus on market was in disorder,and different species had difference on pharmacology and ingredient.Owing to the following questions,this subject took the method of anti-thorm and removing brain edema founding out the efficient part of improving cerebral apoplexy,and accomplishing determination of two active compound,then undertaking extraction and purification of efficient part initially.In this research,thorm and brain edema model of SD rats was set up by the methods of anti-thorm and removing brain edema in clinic.Pharmacodynamic research was taken on Ilexpubesceus of different polarities,and result showed the n-butanol extract of Ilexpubesceus was the efficient part on curing ischemic cerebral paralysis,the n-butanol extract of Ilexpubesceus was separated into four glycoside monomer by using macroporous adsorptive resins,silica column,Rp-C18 column and Sephadex LH-20 column laminar analysis,then structure appraisement was taken on the separated glycoside by several spectroscopy methods such as MS,1H-NMR and 13C-NMR.they are neopubescensin,geniposide,ilexsaponin A.While compoundⅣ's structure appraisement is undertaking now.The result of literature retrieval showed they were separated from Ilexpubesceus on first time.Develop the method of blood disc aggregation in vitro to research the action of anticoagulated blood of the four compounds,result shows they all have the action of anticoagulated blood in different degree.,especially neopubescensin and compoundⅣ, inhibition ratio of anticoagulated blood is 59.6±13.12%and 61.8±11.18%respectively. Assaying of the material was taken by HPLC,and the method of determining neopubescensin and geniposide was established,completing three batchs of medicals' determination.This method is sensitive,accurate,and having good reproduction quality,which can be used to control the quality of medicals of Ilex pubescens.At the same time,finger print analysis of its efficient part on ischemic cerebral paralysis was taken by HPLC and the standard finger printing was established.The research suggested the similarity index between efficient part and standard finger print should be lower than 0.8,by this way the quality of efficient part was controlled,and it would be widely used for its convenience,stability,reproducibility and precision.In addition,the extracting technology of Ilex pubesceus material was studied by using neopubescensin as quality control index,the result showed the best technology was:9 times 60%ethanol taken as extraction solvent,twice decoction of backstreaming with 3 hours each.The cleansing technology study was also taken by mavroporous adsorptive resins to determine the best cleansing technology,the result showed the best technology was:Taking D-101 mavroporous adsorptive resin as medium,washed with distilled water,20%ethanol and 60%ethanol,successively,total glycosides of Ilex pubesceus was enriched in the 60%ethanol elution.In this research,efficient part of Ilexpubesceus on improving ischemic cerebral paralysis was obtained.At the same time,the basic research was accomplished,such as the assaying,finger print analysis,the extraction and cleansing technology,which was meaningful on research of Ilex pubesceus preparation on improving ischemic cerebral paralysis and encouraging the development and utilization of Ilex pubesceus. |
