Objective To establish human oral squamous cell carcinoma cell line ,to ascertain its origin of tissue,and research curcumine's correlated toxicant effect toward squamous cell, to detect a series of bionomics after applying curcumine,such as cell morphous ,growth cycle,the expression of nuclear factor and cyclin .Methods①primary culture: digest method to obtain oral squamous cell carcinoma tissue definited via pathologic means,specimen was digested with 0.25% trypsin and EDTA and dissociated into single cell suspensin,tissues were maintained in PRMI-1640 +10% calf serum at 37℃under 5% CO2;②the phase contrast microscope was used to observe the cell size;③cell group's identify observing the expression of CD44 through immunohistochemical method to ascertain its origin of tissue and cell;④curcumin of different density was admixed with cell of growth index phase,to detect the effect of cell after 24-48 hour ,including cytotoxic effect,the change of morphologic,apoptosis,vegetative cycle and nuclear factor,cyclin .Results Cultured primary cell of oral squamous cell carcinoma successfully.showed polygonal or short spindled;CD44 was positive via immunohistochemical method,combined pathological result to definite it to be squamous cell;curcumin of 5,10,20mg/L obtained cytotoxic effect, cell population in unit area decreased,conjunction disappeared,cell body shrinked or changed round or anomalism,appeared karyopycnosis,cell membrane completed but effervesced and emerged apoptotic body;the cell growth was confined and its ratio increased following drug's density;occurred apoptosis and the phase of G2/M blocked ,the proportion of S and G2/M phase increased ,whereas G0/G1 phase in contrast ,these change was direct correlation with drug density;the positive cell of NF-κB,cyclin D1 reduced and was inverse correlation with drug density.Conclusion①squamous cell carcinoma by primary cultured and passaged in vitro could obtain the category and the characteristic of original tissue,could used to be empirical cell of oral and maxillofacial squamous cell carcinoma .②curcumin inhibited squamous cell's growth,accelerated apoptosis,prevented cell to enter caryocinetic phase;decreaed the expression of NF-κB and cyclin D1 in squamous cell carcinoma process, curcumin could be utilized as a chemopreventive agent toward oral cancer and providing a kind of agent to be safe and effective for the prevention and cure of maxillofacial squamous cell carcinoma.
