Background and objective:At present,there is few investigation about Thyroid Papillary Carcinoma andmany patients who suffered from thyroid carcinoma were always treated by endocrinetherapy.The biotherapy center of NanFang hospital treated many patients whosuffered from thyroid carcinoma with cytokine-induced killer cells(CIK) and IL-2 andobserved good therapeutic effect on these patients. We found that this cellularbiotherapy can lead to the change of thyroid functions in patients.As a result, patientswho underwent these changes can have more good therapeutic effect than otherpatients. Therefore,through in vitro experiments and clinical observation,we try to findout the internal relationship between the biotherapy of CIK and IL-2 and the thyroidfunctions of cancer patients,especially in thyroid carcinoma patients.According to theexpected result,we try to identify the possible mechanism about this phenomenon sothat we can provide the theoretical evidences for the therapy for thyroid carcinomapatients.Material:DMEM(High carbohydratestype),FBS,Insulin,24/96-well plates,Hepes,2mMol/LL-glutamine(Gln),1mMol/L sodium pyruvate,0.25% parenzyme,0.04% ethylenediamine tetracetic acid(EDTA), 0.01Mol/L balanced solution(pH7.4),D-Hank's solution.,Mouse IgG,Rabbit mAb to mouse with HRE Cytokine-inducedkiller cells, Interleukin-2, T3,T4,TSH radio-immunity kits, NAPCO MODEL5410type carbon dioxide electrothermostat, Autoclave sterilizer,Nikon type invertedmicroscope and camera, SIGMA1-13 type centrifugal machine,LSY-â…¡type commoncentrifugal machine, Microamount-sample injector,Fluorescencemicroscope,Electronic analytical balance,Super-clean bench,Eye scissors,Nippers.Methods:1. Tissues of thyroid carcinoma were selectively cut into 1mm3 frompatients.The epithelial cells were cultivated in DMEM with 20%FBS.Thecell of thyroid carcinoma was idengtified by Mouse monoclonal antibody tohuman thyroglobulin.2. The cells were observed by inverted phase contrast microscope andphotographed every day.3. The cultivation and passage of the cells.4. The cells were divided into 7 groups and were stimulated byCytokine-induced killer cells and Interleukin-2.5. The detection of the solution of T3,T4 by radio-immunity kits.6. The detection of proliferation of the cells by Mono-nuclear cell direccytotoxicity assay.7. The selection of clinical patients and the determination of therapeuticstandard with Cytokine-induced killer cells and Interleukin-2.8. The collection of the blood preparations of patients in treatment and thedetection of the solution of T3,T4 and TSH.9. The statistics with SPSS 10.0 software. Conclusion:1. To establish successfully thyroid carcinoma cell model with typicalappearance identified by Mouse mAb to human thyroglobulin.2. The pharmacal research indicated that high solution of IL-2 decrease thesecretary function of thyroid carcinoma cells.However,the median and lowsolution of IL-2 can't.3. CIK and IL-2 could obviously inhibit the proliferation of thyroid carcinoma.4. CIK and IL-2 hadn't any effect on the thyroid function of clinical patients.
