Object: To determine the frequencies of polymorphism in the IL4, ILl3,IL13RA1 gene in Han population of southwest china and to investigate thesusceptibility of the populations to chronic obstructive pulmonary disease(COPD).Methods: To examine the interaction between a IL4, ILl3, IL13RA1 genepolymorphisms and COPD, we carried out a case-control study. 259 patientswith COPD were recruited from southwest china area.245 unrelated,age-matched healthy subjects, who had no known respiratory system illnessacted as control subjects. Genomic DNA was extracted from peripheral bloodwith a Chelex-100 and WB genomic DNA extract kit. Genomic DNA wasused as template for amplification by polymerase chain reaction- RestrictionFragment Length Polymorphism(PCR-RFLP).SPSS was used for statisticalanalysis. X~2 test was used to examine the genotype in COPD and controlgroup. Logistic regression analysis was used to exclude confounding factors and evaluate the effect of smoking or past history on COPD combining withgene polymorphism.Results: 1. The frequencies of these C/T polymorphism of the IL4 promotor-589C/T in COPD group were C/C 0.8%, C/T 21.2%, T/T 78%; Thefrequencies of polymorphic genotypes in control group were C/C 0.8%, C/T14.3%, T/T 84.9%. The allele frequencies of COPD group were C 11.4%, T88.6%. The allele frequencies of control group were C 7.9%, T 92%. The X~2of COPD and control groups were 4.147(P>0.05), 3.377(P>0.05) respectively.The X~2 of polymorphic genotypes in control group and COPD stageⅠ, COPDstageⅠand COPD stageⅡ, COPD stageⅡand COPD stageⅢ, COPD stageⅢand COPD stageⅣwere 0.895(P>0.05), 1.096(P>0.05), 3.782(P>0.05),13.227(P<0.05). Using logistic regression analysis stratified with confoundingfactor. The result between COPD and control were CC: P=0.827 OR=0.8;C/T: P=0.047 OR=1.624; T/T: P=0.827 OR=1.25.2. The frequencies of these A/G polymorphism of the A1398G of IL 13RA1 inCOPD group were A/A 13.1%, A/G 39.0%, G/G 47.9%; The frequencies ofpolymorphic genotypes in control group were A/A 11.0%, A/G 36.7%, G/G52.2%; The allele frequencies of COPD group were A 32.6%, G 67.4%. Theallele frequencies of control group were A 29.4%, G 70.6%. The X~2 of COPDand control groups were 1.11(P>0.05), 1.233(P>0.05)respectively. The X~2 ofpolymorphic genotypes in control group and COPD stageⅠ, COPD stageⅠandCOPD stageⅡ, COPD stageⅡand COPD stageⅢ, COPD stageⅢandCOPD stageⅣwere 0.138(P>0.05), 0.268(P>0.05), 4.722(P>0.05), 4.246(P>0.05). Using logistic regression analysis stratified with confounding factor.The result between COPD and control were A/A: P=0.585 OR=1.177; A/G: P=0.386 OR=1.187; G/G: P=0.585 OR=0.85.3. IL4 promotor -33C/T and ILl3 transposon R130Q polymorphisms can't bedetected in this research.Conclusions: The C/T genotype of IL4 promotor-589C/T may be associatedwith COPD in Han population of southwest china the frequencies ofgenotype were different stageⅢof COPD from stageⅣof COPD. Theremay be no association between the A/G A1398G of IL13RA1 variation ofthe and COPD in Han population of southwest china.
