| Objective: To research the effects of traditional Chinese herb remedies Guan-Xin-Er-Hao(GXRH) and Xiong-Shao(XS) on Hemodynamics in Rats. And, to explore their preliminary Protective Mechanisms of Vascular endothelium.Methods: Modelling duodenal administration and hemodynamic dynamic monitoring in anesthetized rats. Dynamic monitoring of HR,ECG,BP,LVSP,LVEDP,LVDP,PRP and LV dP/dt after pro dosi duodenal administration of XS freeze dried powders (10.2 g/kg, equivalent Chinese herbal medicine remedies 30g/kg) and GXRH freeze dried powders(9.6 g/kg, equivalent Chinese herbal medicine remedies 30g/kg) by PowerLab in 3 hr. Determination of levels NO,NOS,T-AOC,GSH,SOD in serum and myocardial tissue samples, the samples collected at 3 hr in rats after duodenal administration of GXRH or XS freeze dried powders. Simultaneous determination of peoniflorin and ferulic acid in rats after oral administration of XS decoction by HPLC.Results:①Hemodynamic dynamic monitoring the results: XS decreased at 5min-3hr and GXRH decreased at 5-60min on HR (p<0.05 or 0.01); XS and GXRH is insignificant difference on ECGⅡlead J points(p>0.05); XS decreased at 5min-3hr and GXRH at 5-60min on MAP (p<0.05 or 0.01); XS and GXRH on LVSP insignificant difference(p>0.05), but XS decreased at 120min vs GXRH (p<0.05); XS increased at 5-60min and GXRH at 5-30min on LVEDP (p<0.05); XS decreased at 120min on LVDP vs GXRH (p>0.05); XS decreased at 15min-3hr and GXRH decreased at 30-60min on PRP(p<0.05或0.01); XS and GXRH is insignificant difference on LV dP/dt max(p>0.05); XS increased at 5-15min and 60min-3hr and GXRH increased at 10-90min on LV dP/dt mix (p<0.05). XS and GXRH moved the lowest points to top-right on their pressure correlation loop (PCL).②GXRH and XS vs NS groups: GXRH and XS on NO increased in serum samples(p<0.05), on T-NOS increased in serum and myocardial tissue samples(p<0.01 or 0.05), on iNOS decreased in serum samples(p<0.01 or 0.05) and myocardial tissue samples(p>0.05), on T-AOC and GSH increased in serum and myocardial tissue samples(p<0.05), on T-SOD and CuZn-SOD decreased in serum samples(p<0.01), the samples collected at 3 hr in rats after duodenal administration of GXRH or XS freeze dried powders.③Simultaneous determined preliminary peoniflorin and ferulic acid in plasms after oral administration of XS freeze dried powders in rats by HPLC.Conclusion: traditional Chinese herb remedies GXRH and XS have the efficacy of negative chronotropic property and depressurization. Prompting GXRH and XS have the indirect action of arteriectasis, decrease myocardial oxygen consumption(MOC) and increase coronary blood flow(CBF) in Anesthetized Rats. Mechanisms of depressurization lie in the efficacy of negative chronotropic property and endothelial vasodilatation. They have the effect of antioxidation and accommodate redox homeostasis in vivo, therefore, protection on Vascular endothelium. established preliminary the active components of XS remedies is peoniflorin and ferulic acid in rats. |
