| Objective To detect the contents of TGF-β1 on corneal grafts, and to study the effect of TGF-β1 during the penetrating keratoplasty rejection.Methods Penetrating keratoplasty was proformed orthotopically from Wistar rats to SD rats. The rats include three groups: experimental group(TGF-beta1 humor ocular), control group(Chloroptic), and self group(Chloroptic). The appearance of rejection were studied and recorded by the biomicroscope examination. A half of whole corneas of each experimental groups were enucleated at random at first week, second week, third week, first month, and second month postoperatively. The immunohistochemistry combined computer-assisted image analysis was adopted to examine the expression and distribution of TGF-beta1 and IL-1βon corneal grafts .Results 1. The mean survival time of transplants increased significantly in the experimental groups than the control group(P<0.01). 2. The level of expression of TGF-beta1 in all experimental groups was higher than this in the control(P<0.01). 3. The level of IL-1βin all experimental groups was markedly lower than this in the control group at different periods postoperative(P<0.05).Conclusion The keratoplasty graft rejection can be reduced and mean survival time can be prolonged by TGF-beta1, which inhibits the expressions of IL-1β.PartⅡ: The experimental study of the effect of IL-6, IL-8 on penetrating keratoplasty rejection in ratsObjective To detect the contents of IL-6, IL-8 in peripheral blood during the penetrating keratoplasty rejection, to study the dependablity of IL-6, IL-8 and TGF-beta1, and to analysis their roles in the corneal allograft rejection.Methods Penetrating keratoplasty was proformed orthotopically from Wistar rats to SD rats and from Wistar rats to Wistar rats. The rats include three groups: experimental group(TGF-beta1 humor ocular), control group(Chloroptic), and self group(Chloroptic). ELISA assay was used to detect the contents of IL-6, IL-8 in the peripheral blood after the rats are sacrificed at different times.Results 1. The level of expression of IL-6, IL-8 in all experimental groups was markedly lower than this in the control groups and self groups at different periods postoperative(P<0.01).Conclusion 1. IL-6, IL-8 are important factors to start corneal graft rejection. 2. The expression of IL-2mRNA in experimental groups was markedly lower than this in the control groups and self groups at different periods postoperative(P<0.05).Conclusion IL-2 have a very important effect in corneal graft rejection, and its levels can be reduced by TGF-β1, which can be markedly prolong the corneal graft's mean survival time... |
