Objectives: To evaluate the presence of DC in small airways of COPDmodel rats. And to observe the effects of inhaled budesonide andipratropium bromide on the model. Methods: Sixty male SD rats were randomdivided into four group: control group(A group),COPD model group(Bgroup),budesonide-treatment group(C group)and ipratropiumbromide-treatment group(D group). Rats in B,C and D group were exposedto the tobacco smoke of 4 cigarettes twice a day with 30 minutes each time.Lipopolysaccharide (LPS) 200ug was instilled intracheal to these ratstwice at the first day and the 14th day. The control groups(n=15) wereexposed to air. C group rats received inhalation of Pulmicort 2 mg (4 ml)since the second week and D group rats received inhalation of Atrovent1mg (4ml) at the same time. At the 31th day we intraperitoneal injectedpentobarbital 40mg/kg to anesthetize the rats, cut the belly open, andexecuted them by blood letting from abdominal aorta. Quickly opened thechest and isolated inferior-median lobe of right lung and inferior lobeof left lung. The inferior-median lobe of right lungs were fixed by 10%formalin, paraffin imbedded, HE stained. Observe pathological change in lungs of rats with light microscope. At the same time, inferior lobe ofleft lung were wraped up with tinfoil, freezen with liquid nitrogen.Investigated the distribution of DCs in each group byimmunohistochemistry with a rat-DC-specific monoclonal antibody, OX62.Measurements and Main Results: COPD group and drug inhalation groupexpressed lassitude, hair tarn, less of weight's increase than controlgroup, locomotor activity declining, coughing, increasing of excretion inrespiratory passage. HE staining expressed the character pathobiologychanger of COPD. There is a significant increase in DC number in lungs ofCOPD model group rats compared with the control group rats(p<0.01). DCnumber in budesonide-treatment group also increases, but less than it inmodel group(p<0.01).There are no different between the ipratropiumbromide-treatment theatment group with the model group (P>0.05).Conclusions: There is significant increase in DC number in lungs of COPDmodel group than control group. Giving budesonide-treatment will reducethis change.
