| Objective: In this study, we investigated the protein expression of MMP-9, TIMP-1, Phospho-c-Jun, and MMP-9/TIMP-1 in peripheral blood mononuclear cells of patients with coronary heart disease, and the severity of coronary artery by coronary angiography to investigate their possible regulations and correlations .Methods: we selected 55 patients enrolled in our hospital from October 2005 to June 2006, and were diagnosised as coronary heart disease. All of them were underwent an immediately or selectively Coronary angiography (CAG). The protein expression ofMMP-9,TIMP-1 and Phospho-c-Jun in peripheral blood mononuclear cells were measured by western-blot in 24 patients with ACS(8 with AMI and 16 with US), 16 with SA and 15 control subjects. Coronary angiograms were scored according Gensini integration, analysis the relationship between MMP-9, TIMP-1, Phospho-c-Jun, MMP-9/TIMP-1 and Gensini integration by linear correlation.Result:⑴T he protein expression of MMP-9, TIMP-1 and the ratio of MMP-9/TIMP-1 were significantly increased in ACS group than that in SA group and control group(all, P〈0.001),but there was no statistic differences between the later two groups(P>0.05);⑵The protein expression of Phospho-c-Jun was significantly increased in ACS group and SA group than that in control group(respectively, P〈0.001,P〈0.05), The former two groups also have significant statistic differences(P〈0.001);⑶The protein expression of MMP-9, Phospho-c-Jun and the ratio of MMP-9/TIMP-1 were positively correlated with Gensini integration(respectively r=0.433, r=0.476 and r=0.457,all P<0.01);But there was no correlation within TIMP-1 protein expression and Gensini integration;⑷The protein expression of MMP-9 was greatly correlated with phospho-c-Jun in patients with coronary heart disease(r=0.839,P<0.001).Conclusions:⑴The protein expression of MMP-9 and the ratio of MMP-9/TIMP-1 were significantly increased but imbalanced in the patients with ACS, which suggested a prediction of more vulnerable plaque in ACS .⑵The protein expression of MMP-9, phospho-c-Jun and the ratio of MMP-9/TIMP-1 were positively correlated with Gensini integration, which suggested an implication of the severity of coronary artery.⑶T he activation of transcription factor c-Jun may contribute to the expression of MMP-9 during the pathogenesis of artherosclerosis. |
