Objective:1,To research the changes of the Plasma Lysophosphatidic Acid(LPA) Concentration and Platelet Aggregation Maximum(PAGM)in Experimental Rats with acute cerebral thrombosis;2,To study the effects and mechanisms of only using dipyridamole before acute cerebral thrombosis in rats on plasma Lysophosphatidic Acid(LPA)Concentration and Platelet Aggregation Maximum(PAGM);3,To study the effects and mechanisms of only using dipyridamole before acute cerebral thrombosis in rats on brain tissue changes.Methods:Acute cerebral thrombosis rats model were produced through injecting thrombin into the internal carotid artery(ICA)by introducing a catheter.Thirty Wistar rats were randomly divided into five groups(Normal control group,Sham-operation group,Oper-model group, dipyridamole pretreatment control group and NS pretreatment control group),each had six rats. Observe the changes of plasma LPA and PAGM of all the rats in each group,and the degree of the impairment of neural function was scored,the histological changes were evaluated and cerebral infarction volume were calculated.Using Abs-ratios detect LPA;Using turbidmetric method to measure PAGM by ADP antileptic induced.Results:The concentration of plasma LPA and PAGM in Normal control group were low(2.32±0.28μmol/l and 52.00±1.59%respectively)and in rats of Oper-model group were high(12.58±1.57μmol/l and 82.30±2.15%respectively).The concentration of plasma LPA and PAGM had significant differences between the two groups(P<0.05).The rats in dipyridamole pretreatment control group were instilled dipyridamole into the stomach before the surgery,then the concentration of plasma LPA and PAGM decreased markedly(4.35±0.75μmol/l and 63.92±2.34%respectively)and the concentration of plasma LPA and PAGM in rats of the NS pretreatment control group were still high(13.08±1.79μmol/l and 84.38±2.31%respectively). Between NS pretreatment control group and Oper-model group they had significant differences (P<0.05).After acute cerebral thrombosis in rats,using light microscopy:the brain histological Changes were normal in Normal control group and Sham-operation group,but in Oper-model group and NS pretreatment control group brain tissue had hydropsia and ischemic changes obviously.In dipyridamole pretreatment control group brain tissue had hydropsia slightly.Normal tissue were red,but infarctus tissue were white.Normal control group and Sham-operation group were normal,but Oper-model group and NS pretreatment control group had major white infarctus tissue;dipyridamole pretreatment control group had micro-infarctus tissue.Between Oper-model group and Normal control group,Sham-operation group they had significant differences(P<0.001),Between dipyridamole pretreatment control group and Oper-model group they also had significant differences(P<0.01).Conclusion:The concentration of plasma LPA and PAGM increased in Oper-model rats with acute cerebral thrombosis,and had ischemic changes obviously,in brain tissue,that indicates the experimental rats models with acute cerebral thrombosis in our experiment were successful; Instilling dipyridamole into the stomach of the rats could decreased the concentration of plasma LPA and at the same time inhibited the increase of plasma PAGM obviously,and decreased ischemic changes in brain tissue,so that dipyridamole could prevent ischemic cerebrovascular disorders.
