| Objective:To investigate induction of the expression of humanβ-defensin-2(hBD-2) mRNA,β-defensin-3(hBD-3)mRNA by TNF-α,IFN-γin human keratinocyte cell lines (Hacat cells),and to understand the basic regulation of induction of the expression of human hBD,which will provide useful information to the further research of hBD.Methods:Cultured Hacat cells was divided into eight groups:contrast of 12h,contrast of 24h,stimulated by TNF-αfor 12h,stimulated by TNF-αfor 24h,stimulated by IFN-γfor 12h,stimulated by IFN-γfor 24h,stimulated by TNF-αand IFN-γfor 12h,stimulated by TNF-αand IFN-γfor 24h,Cultured Hacat cells were stimulated with 40ng/ml TNF-α,40ng/ml IFN-γ,40ng/ml TNF-αand 40ng/ml IFN-γfor 12h or for 24h,The expression of hBD-2mRNA,hBD-3 mRNA was detected by reverse transcription polymerase chain reaction (RT—PCR).Results:HBD-2 mRNA,hBD3- mRNA expression was few in Hacat cells;HBD-2 mRNA,hBD3-mRNA expression was up-regulated by TNF-αin Hacat cells,significantly,The hBD-2 mRNA,hBD3-mRNA expression at 12h post higher than 24h post;HBD-3 mRNA expression was up-regulated by IFN-γin Hacat cells,significantly,The hBD-3 mRNA expression at 12h post higher than 24h post,but hBD2-mRNA expression wasn't up-regulated by IFN-γin Hacat cells;There are the interaction in the three faction of the time,TNF-α,IFN-γwhen hBD-2 mRNA,hBD3-mRNA expression was up-regulated by TNF-αand IFN-γ.Conclusions:This report demonstrate the presence of hBD-2,Hbd-3 in human keratinocytes and capacity of human keratinocytes to produce hBD-2,hBD-3 in response to TNF-αand to produce hBD-3 in response to IFN-γ,and these courses of induction is time dependent,in addition,there are the interaction in the three faction of the time,TNF-α,IFN-γwhen hBD-2 mRNA,hBD3-mRNA expression was up-regulated by TNF-αand IFN-γ. Release of hBD-2,hBD-3 by keratinocytes in response to TNF-α,IFN-γmay contribute to resisting infections and resisting tumors and the host defense responses of the skin. |
