The Experimental Study Of The Role Of NRP-1in Promoting The Astragalus HaCaT Cell Proliferation

Objective:To research the influence of Astragalus to normal HaCaT cells proliferation and NRP-1expression and to transfected with NRP-1shRNA plasmids HaCaT cells proliferation and NRP-1expression, in order to explore the possible mechanism of astragalus in promoting the healing of varicose ulcer.Methods:Using RNA interference technology to transfected NRP-1shRNA plasmid into HaCaT cells. Observed the level of HaCaT cell proliferation after transfection and Astragalus extract raction after transfection with MTT method, and c Observed the level of HaCaT cell proliferation after transfection and Astragalus extract raction after transfection with RT-PCR method.Results:NRP-1shRNA transfection can effectively silence NRP-1expression in HaCaT cells and reduced cell proliferation levels. After NRP-1shRNA transfection, the role of Astragalus extract in HaCaT cell proliferation and promote the role of NRP-1expression is suppressedConclusion:Astragalus extract promoting proliferation of HaCaT cells may be related to’ pro-HaCaT cells NRP-1expression.
