Objective: To study the expressions of heat shock protein 27(Hsp27) and p38 mitogen-activated protein kinase(MAPK), and the effects of flunarizine on the expressions of Hsp27 and p38MAPK in brain tissue following transient cerebral ischemic reperfusion in gerbils.Methods: A cerebral ischemia-reperfusion model in gerbils was established by clamping both common carotids. Fifty gerbils were randomly divided into: normal control group, sham operation group, ischemia-reperfusion(IR) group, flunarizine intervention group . Gerbils were fed with flunarizine(20mg/ kg) the day before experiment in flunarizine intervention group .The expressions of Hsp27 and p38MAPK in brain tissue were detected by using immunohistochemistry technique at the 6th hours, 1st day, 3rd and 7th day after IR. Experimental results were analyzed with Spss13.0.Results: Experimental results revealed that Hsp27 were expressed in neurons and gliacytes of the brain tissue and p38MAPK was expressed in gliacytes in 50 gerbils. There were few expressions of Hsp27 and p38MAPK in neurons and gliacytes both normal control group of 5 gerbils and sham operation group of 5 gerbils, and there was no significant difference between the two groups (P>0.05). Compared to that of normal control group, the expressions of Hsp27 and p38MAPK in neurons gliacytes of ischemia-reperfusion gerbils were evidently increased at the 6th hour,lst day, 3rd day and 7th day,especially at the 3rd day ( p < 0.05 ) . Compared to IR group, the expressions of Hsp27 and p38MAPK in neurons and gliacytes of gerbils in flunarizine intervention group were evidently decreased at the 6th hour,lst day, 3rd day and 7th day ( p < 0.05 ) .The expressions of Hsp27 and p38MAPK in neurons and gliacytes of gerbils in normal control group , ischemia-reperfusion group and flunarizine intervention group were positive correlation, the coefficient correlation is 0.978 ( p < 0.05 )Conclusions:1. The expressions of Hsp27 and p38MAPK in neurons and gliacytes of gerbils were evidently increased following cerebral ischemia-reperfusion.2. The damage of brain tissue in Flunarizine intervention groups became moderate and the expressions of Hsp27 and p38MAPK decreased than those of the expressions in the IR group.3. Expressions of Hsp27 and p38MAPK in neurons and gliacytes of gerbils were positive correlation, this phenomenon may show that HSP27 is one of the downstream in p38MAPK signal transduction pathway.
