| Objective: The effects of Liqi Huoxue and Qingre fomula and its every separated formulas (Q,L,H,QL,QH,LH) on PDGFRβ-PLC-γ1-PKCαsignal transduction in human gastric adenocarcinoma cell line BGC823 were observed to investigate the therapeutic mechanism of clearing heat,regulate qi, invigorating the blood and to discuss the relationship between heat toxin, qi stagnation ,blood stasis and PDGFRβ-PLC-γ1-PKCαsignal transduction in human gastric adenocarcinoma cells.Methods: After BGC823 cells were treated with Qingre Liqi and Huoxue fomula and its every separated formulas (Q,L,H,QL,QH, LH),the morphological change of cell was observed under a inverted microscope, the best formula was screened by orthogonal tests and the expression of PDGFRβ,PLC-γ1 and PKCαwere measured by Western blot analysis.Results:①W and its every separated formulas can significantly inhibit BGC823 cell growth and cause the different morphological changes within each group, The cells treated by Q show very rough cytoplasm, being filled with a lot of accumulation, and vacuole occurrence in matrix .The cells treated by L show ribbon shape, cell shrinkage, wide cell-cell space and rough cytoplasm.The cells treated by H show shrinkage, transmittance decrease, reticulum occurrenc in the matrix among cells.The cells treated by W and others separated formulas show very similar to the cells in Q and L groups,but there were more cell fragments in media.②W and its every separated formulas can all significantly inhibit BGC823 cell growth,in which W is the strongest and dose-dependent.③PDGFRβ,PLC-γ1,PKCαwere overexpressed in human gastric adecacinoma cell line BGC823.④When the cells were treated with IC50 dosage, the protein expression level of PDGFRβ,PLC-γ1 and PKCαin the cells decreased, the result is: W> LH > QH > H > QL > L >Q。Conclusions:①W and its every separated formulas significantly inhibit the growth of BGC823 cell. In which W is the strongest that shows Q, L and H have synergistic effects.②There is close relationship between cell growth and PDGFRβ-PLC-γ1-PKCαsignal transduction in BGC823 cell.③W and its every separated formulas down-regulate the protein expression level of PDGFRβ-PLC-γ1-PKCαsignal transduction.They may inhibit the growth of BGC823 cells through inhibiting PDGFRβ-PLC-γ1-PKCαsignal transduction pathway.④heat toxin, qi stagnation ,blood stasis had close relationship with the marked increment of PDGFRβ-PLC-γ1-PKCαsignal molecular expression and transduction in the phathogenesis of human adenocarcinoma.⑤It should be emphasised that clearing heat,regulate qi, invigorating the blood circulation is a very important method in treating human gastric cancer. |
