World-wide people are familiar-with influenza, the disease caused by the orthomyxoviruses, The family orthomyxoviridae comprises influenza A, B, and C viruses. Epidemics which must have been caused by influenza viruses have been lasted for 2000 year. Typically there is a sudden appearance of cases of respiratory disease, these last for several weeks and then suddenly cease. Epidemics of influenza lead to lots of losses, so people pay more attention to seek the methods to treat and prevent influenza.Arctium is a common Chinese medicine which is often applied to treat such diseases as influenza, acute tonsillitis, herpes simplex and so on that caused by viruses clinically. Experiments in vivo showed that Arctigenin at the concentration of 100μg/kg and 10μg/kg for the intranasal treatment markedly inhibited the lung consolidation of mice pneumonia caused by the infection of influenza virus. Arctigenin at the concentration of 100μg/kg prolonged the survival time of mice infected with influenza virus.The paper has mainly researched two aspects:(1) The effect of anti-influenza virus of Arctigenin Compound in vivo;(2) Research the antiviral mechanism of Arctigenin Compound from the view of interferon inducing in mice on the basis of anti-viral effect of Arctigenin CompoundPartⅠ: Literature OverviewThe author has reviewed the ancient and up-date data and knowledge especially treatment of influenza. Success came in 1933 when sir Christ Pber Andrewes and others isolated influenza A virus. Continued isolation studies and analysis of isolates have given an understanding of how the epidemic behavior relates to change in the virus. At the present, some medicines which have been used to treat influenza, such as amanltadine, rimantadine and ribavirin, can effectively inhibit the influenza virus, but the applying of them is limited because of their side-effects. So far, this infectious disease has been not effectively controlled. But the data showed that traditional Chinese medicine (TCM) had played a more and more important role in treatment of influenza. So, it is necessary to further study the antiviral mechanism of TCM.PartⅡ: Laboratorial studyObjective:To Prove the antiviral effect and its mechanism of Arctigenin Compound through laboratorial study with modem medical methods, and to offers a scientific foundation for the applying of Arctigenin Compound on influenza.Methods:In vivo, the influences on lung index, death rate of mice infected by influenza virus was observed in the mice infected with virus from nose. And the effects of Arctigenin Compound on interferon inducing in normal mice were observed, and then the serum interferon was determined after a single Arctigenin Compound administration.Results:In vivo, Arctigenin Compound could improve the mouse Pneumonia caused by influenza virus(comparison with model control group, P<0.05); It could prolong the life span of mouse infected with Pneumonia by Influenza virus A strain FM1. The interferon contents were detected in 18h after Arctigenin Compound administration.Conclusion:Arctigenin Compound inhibited the proliferation of influenza virus A and improved the symptom of mouse Pneumonia caused by influenza virus, and reduced the mortality rate of Intranasal infected, prolonging their lives. Based upon the results it appears that the Arctigenin Compound may be valuable in treating influenza。And, Arctigenin Compound can exert enhancing effects on interferon induction in vivo.The antiviral effects of Arctigenin Compound exert enhancing effects on interferon induction in vivo in vivo provided scientific experimental basis for treatment on influenza in clinic.
