According to the modern medical theory, Yin-deficiency syndrome is a basic physiological process and syndrome that caused by the pathogenic factors. There must be a series of pathologic and physiological reaction when it develops. That is to say, there are comprehensive and thorough pathologic changes when the Yin-deficiency syndrome occurs. The structure and function of every system, organ, tissue and/or cell have different changes (primary change and/or secondary change).Based on the principle of "structure and function unify" in modern medicine theory. Prof. WeiXi Shen proposes that the essence of Yin-deficiency syndrome can be cytokines, as its pathogens can be due to the perturbation of cytokines networks characterized by the relative elevation of the gene expression and the biology activity of IL-1, TNF and so on, which results in the turbulence of cytokines networks. The entire biological phenomenon and the course of disease caused by cytokines are the final results of a series of complicated control in the cytokines networks. At present, the study conclusion that the essence of Yin-deficiency is cytokines remain only a theoretical model and theoretical frame. If we want to uncover the specific content about the changes of the cytokines networks when Yin-deficiency syndrome occurs, signal transduction pathway, the regulation mechanism of nuclear transcription factor to inflammatory cytokines. It can be resolved only through empirical study. On one hand, the essence of Yin-deficiency syndrome is complicated; the tools available to clarify its regulation also need to be advanced. cDNA micro-array technology is a powerful descriptive method for determining the expression profile of hundreds to thousands of genes. Since then it has been used successfully in various areas of gene function research. Large-scale gene expression profiling using cDNA arrays has become one of the most fruitful methods for characterizing the physiological and pathological process. Using this technique to look for the difference of expressed genes in the essence of Yin-deficiency syndrome will be helpful for the analysis of the mechanism and clinical treatmentExperiment studyMethods: To select 20 cases of lung cancer patients with Yin-deficiency syndrome and 20 cases of non-Yin-deficiency syndrome control, They were divided in two groups: Yin-deficiency group and non-Yin-deficiency control group. In the Yin-deficiency group, 11 patients are female and 9 patients are male, whose age is from 41 to 70 years old. In the non-Yin-deficiency control group, 10 patients are female and 10 patients are male, whose age is from 37 to 70 years old. All the samples were resected from patients who had been diagnosed and operated. These samples were detected through SABC method of immunohistochemistry. Results: the expression level of MAPK NF-κB AP-1 protein in the lung cancer patients with Yin-deficiency syndrome group is higher than that of in the non-Yin-deficiency group(P<0.05).Methods: To select 3 cases of cancer patients with Yin-deficiency syndrome and that of 3 cases of non-Yin-deficiency control. They are all female with lung cancer, the patient with Yin-deficiency is 64 years old and the non-Yin-deficiency patient is 60 years old. The two patients of colon carcinoma are all male, the patient with Yin-deficiency is 58 years old and the non-Yin-deficiency patient is 55 years old. The gastric cancer patient with Yin-deficiency is 61 years old and the non-Yin-deficiency patient is 58 years old. They are all female. Both mRNA from the disease and normal sample were reversely transcribed to cDNA, the following is the hybridization of gene chip, high-stringent washing and analysis, and then hybridization the expression profile chip according to the result of the test gene chip. The chip was scanned by the high resolution scanner and then showed the differences between the two groups. Results: The expression signal value of MAPK,NF-κB,AP-1 protein in Yin-deficiency syndrome group is higher than that of in non-Yin-deficiency control group.Conclusion:After analyzing the theory of the essence of Yin-deficiency and the regulation mechanism of the MAPK, NF-κB and AP-1 to the inflammatory cytokines, and combining all our experiment results, we can draw a preliminary conclusion:1. In the non-cancer cells the expression level of MAPK NF-κB AP-1 protein has increased in lung cancer patients with Yin-deficiency syndrome.2. The expression level of MAPK NF-κB AP-1 gene has changed in cancer patients with Yin-deficiency syndrome. There is something to do with Yin-deficiency syndrome.3. MAPK, NF-κB and AP-1 take part in the signal transcription in the state of Yin-deficiency syndrome which are probably the mechanism of inflammatory cytokines in Yin-deficiency syndrome.
