The modernization of Traditional Chinese Medicine has become the main direction with the development of TCM. It has been believed that the vital activity is related with the cell signal transduction pathway. From the above, we can seen that it is important to discuss the essence of TCM by doing some research on the changing tendency of signal transduction system.Some researches have been done on the etiology and pathogenesis of syndrome of spleen deficiency during the past period. Now, the research on the essence of the syndrome of spleen deficiency is begin to be concentrated on the signal transduction system, which is only on the some isolated factors. And the research on the aim of signal transduction is much less.This research is to investigate the changes of signal transduction in the syndrome of spleen deficiency by detecting the activity of PKC and MAPK of the heart, liver, spleen, lung, kidney, jejunum and brain. It is to testify the essence of the syndrome of spleen deficiency and the mechanism of Chinese herbs of replenishing spleen.The main content of research includes:1. The changes of expression of PKC activity and subtypes in the rats heart, lung and jejunum tissue: The PKC activity in the cytoplasm, cell membrane and the PKC subtypes(PKC , PKC - I and PKC ) have been detected in the spleen Yin deficiency and spleen Yang deficiency rats by substrate phosphorylation and western blotting methods.2. The effect on the PKC activity and subtypes expression in the rats heart, lung and jejunum tissue of Chinese herb of replenishing spleen: It is to observe the effect of PKC activity and subtypes expression of the heart, lung and jejunum tissue by the treatment of spleen Qi deficiency, Yin deficiency, Yang deficiency with professor Li Dexin's herbs.3. The changes of the rats heart, kidney and brain PKC activity and MAPK/ERK1,2 subtype expression in the spleen deficiency: The changes of MAPK activity of the heart, the liver and the brain of the rats with spleen Qi deficiency, spleen Yang deficiency and spleen Tin deficiency were detected by immunoprecipitation and western blotting. And theexpression of ERK1 and ERK2 of the heart and brain of the rats with the same syndrome was detected by the same method.4. The effect of MAPK activity and PKC activity and ERK1 and ERK2 subtypes of the heart, lung and jejunum tissue by the Chinese herbs of replenishing spleen with the rats spleen deficiency.5. The relation between the changes of PKC activity and the changes of MAPK activity of the heart tissue with the rats spleen deficiency.6. The effect of Chinese herbs of replenishing spleen on the relation between the changes of PKC activity and the changes of MAPK activity of the heart tissue with the rats spleen deficiency.It shows that there existed the changes of PKC activity and MAPK activity of different tissues with the rats spleen deficiency and the treatment of the Chinese herbs of replenishing spleen were obviously effective in this syndrome.1. The changes of PKC activity in the different tissue with the rats syndrome(1) The heart tissue: The PKC activity in the cytoplasm of the rats heart was increased obviously with the three types of spleen deficiency syndrome and there was no obvious difference of the increasing amount. The PKC activity in the cell membrane of the rats heart was increased obviously with spleen Qi deficiency and spleen Yin deficiency. The total PKC activity in the cells of the rats heart was increased obviously with the three types of spleen deficiency syndrome and there was no obvious difference of the increasing amount. The PKC activity in the cytoplasm was more than that in the cell membrane in the normal rats heart. But there is no difference of PKC activity between the cytoplasm and the cell membrane in the rats with the spleen deficiency syndrome.(2) The lung tissue: The PKC activity in the cytoplasm of the rats lung was decreased obviously with spleen Yang deficiency and spleen Yin deficiency.
