Objective:In order to investigate the pathogenetic miachnism of spontaneous abortion and to look for available therapetutics theroy,we research apoptosis and expression of Bcl-2 and Bax in deciduas tissue of spontaneous abortion mouse models and normal pregnancy mouse models respectively.Methods:The abortion-prone CBA/JXDBA/2 matings were established as the model of spontaneous abortion and The nonabortion-prone CBA/JXBALB/c matings were used as the model of normal pregnancy.SABC was used to detect the expression of Bcl-2 and Bax in trophoblast and decidua tissue of spontaneous abortion and normal pregnancy model at day 13 of gestation.Medical color-image measure system for immune-histochemistry was used for semiquantitative analysis.Apoptosis was detected by using DNA in situ terminal deoxynuclotidyl transferas-mediated dUTP-biotin nick ending labeling(TUNEL)Results:(1)AI of trophoblast and decidua tissue in spontaneous abortion model was significantly higher than that of normal pregnancy model(P<0.01).(2)The expression of fasL in trophoblast of spontaneous abortion model was lower than that of normal pregnancy model(P<0.01).The expression of fas had no significant difference between two models in trophoblast(P>0.05).The expression of fasL in decidua of spontaneous abortion model was higher than that of normal pregnancy model(P<0.05).The expression of fas had no significant difference between two models in decidua,too.(P>0.05)(3)The expression of Bax in the trophoblast of spontaneous abortion model was higher than that of normal pregnancy model(P<0.05);The expression of Bcl-2 had no significant difference between two model in trophoblast(P>0.05).The expression of Bax in the decidua of spontaneous abortion model was higher than that of normal pregnancy model(P<0.05),too;the expression of Bcl-2 was lower than that of normal pregnancy model(P<0.01).Conclusions:To break the regulation of decidual tissue apoptosis may directly or indirectly results in spontaneous abortion during early gestation.The channel of Bcl-2/Bax systems may be one of the mechanisms of spontaneous abortion deciduas apoptosis.
