ObjectiveTo investigate the effect of the insulin-like growth factor(IGF-1) on the detrusor ofbladder.Detrusor muscle weakness is a common disease in Urinary Surgery,which canderive from the urinary retention that is the result of the neurogenic bladder,diabetesand benign prostate hyperplasia.To improve the myotility of the detrusor of bladder isthe effective method of treating the detrusor muscle weakness.Now most of researchersmajor in improving the regulation of nervous pathway to increase the myotility of thedetrusor muscle. And in recent years the pathway of the IGF-1 becomes more and moreattractive to some researchers because it exists so widely that it can regulate everypathway in the body including a series of regulators and their correspondingligands.This article observes the levels of the IGF-1 in the detrusor muscle of thepatients who suffer from the detrusor muscle weakness from two aspects of mRNA andproteins by RT-PCR and Western blot.And to ensure whether IGF-1 play a role in thepathogenesis of the detrusor muscle weakness,moreover to provide the theoretical basisfor the clinical pathogenesis and improving the myotility of the detrusor muscle byregulating the expressions of IGF-1.MethodTo take 20 patients with BPH as the study group who undergoes the bladderdecision operation and based on literatures to ascertain the existence of UAD by theurethral canal linear passive resistence correlation coefficients in uro-kineticsexamination.Correspondingly,to take 20 patients with BPH as the control group whoundergoes the bladder decision operation but exclude UAD.By RT-PCR and Westernblot to detect the level of IGF-1 in the bladder smooth muscle from the level of mRNAand protein. ResultThis article divide the detrusor muscle samples into 2 groups:BPH withUAD(study group) and BPH without UAD(control group).To detect the level of IGF-1mRNA by RT-PCR and take(?)-actin as the internal reference:the respective densitypercentage of the study group and the control group is 71.6% (?) 19.0% and135.5% (?)34.7% over(?)-actin.And the level of IGF-1 mRNA of the study groupdecreases significantly,and there is a significant difference between the study group andthe control group(P<0.01,n=20) . To detect the level of IGF-1protein by Westernblot:both groups show a strap in the same site,but there is a significant differencebetween 2 groups in contents,BPH with UAD 41668.27 (?) 4688.02 while BPH withoutUADl38585.00(?)5728.76(P<0.01,n=20) .ConclusionThe decreasement of the expression of IGF-1 may play a role in the pathogenesisofUAD in BPH.
