| Objective Through the establishment of benign prostatic hyperplasia induced rat bladder outlet partial obstruction model, we try to detect the weight and structure of bladder as well as the expression of growth factors (transforming growth factor betal, basic fibroblast growth factor), in order to explore the effect and mechanism of Bushen Huoxue recipe on the pathological changes of bladder detrusor musculature caused by bladder outlet partial obstruction.Methods 100 of the 3-month-old male Wistar rats were randomly divided into five groups, 20 rats in each group, in addition to the control group, the remaining four groups of rats were castrated, eighth days after operation daily injection of testosterone propionate and were given normal saline, Bushen Huoxue recipe, finasteride, tamsulosin by gavage, the rats of control group were given normal saline. After 30 days, the rats were sacrificed, rapid remove the prostate and bladder, Calculat prostate and bladder organ index; paraffin section and HE staining is used to observe the changes of bladder smooth muscle, fibrous connective tissue, in IHC immunohistochemical method detect bladder tissue collagen â…¢ and collagen â… level and distribution and collagen â…¢/â… ratio; by RT-qPCR (real-time fluorescent quantitative polymerase chain reverse reaction)detect the expression level of TGF β1, bFGF mRNA of the bladder tissue.Results1. The comparison of each rat prostate, bladder wet weight and organ index:Compared with control group, the body weight of model group, tamsulosin group and traditional Chinese medicine group, finasteride group were decreased significantly (P< 0.01), prostate wet weight and organ index was higher significantly (P< 0.01); bladder wet weight and organ index increased significantly (P< 0.05), in which the drug intervention groups bladder wet weight and organ index were lower than that in the model group, Chinese medicine group, non finasteride group compared with the model group with significant difference (P< 0.01).2. The pathological changes of prostate, bladder under light microscope and electron microscope 1) The pathological changes of prostate tissue under the light microscope:Compared with the control group, under the light microscope the prostate glands of model group rats showed more dense, glandular cavities of different sizes, serrated, different secretions, ranging high columnar epithelial cells, the majority of papillary protruding into the cavity.2) The pathological changes of bladder tissue under the light microscope:Compared with the control group, the modeling groups detrusor hypertrophy, the submucosa and muscular bundles of collagen deposition increased, muscle thickening and uneven, the nucleus is slightly larger and darker. The model rats pathological changes more significantly.3) The results of detecting bladder tissue collagen â… and collagen â…¢ by Immunohistochemistry: collagen I expression difference is not statistically significant (P> 0.05); the expression of collagen III, modeling groups were significantly higher than that in control group (P< 0.05), while the traditional Chinese medicine group, finasteride group were significantly lower than that in model group (P< 0.01); each model groups collagen â…¢/collagen I ratio compared with the control group increased to varying degrees, model group, tamsulosin group with the control group, with significant difference (P< 0.01), Chinese medicine group, non finasteride group was significantly lower than that of model group (P< 0.01), compared with the control group were elevated, but no significant difference (P> 0.05).3. The expression levels of TGFβ1, bFGF mRNA in Bladder tissue1) The expression level of TGF β1 mRNA in each group were compared with the control group is slightly lower, but the differences were not statistically significant (P> 0.05), the difference between the groups to make die no statistical significance (P> 0.05).2) The bFGF mRNA in the expression mode in each group were higher than those in the control group, with significant difference (P< 0.05), the expression level of drug intervention group were lower than model group, model and traditional Chinese medicine group, model plus finasteride group was significantly lower than that of model group (P< 0.01).Conclusion Prostatic hyperplasia can lead to the rat bladder outlet section obstruction, bladder smooth muscle hyperplasia, hypertrophy, type â…¢ collagen deposition increased significantly, increased expression of bFGF mRNA. The early intervention of Bushen Huoxue Recipe can effectively inhibit expression of bFGF mRNA, reduce the pathological changes of the bladder smooth muscle, inhibit excessive deposition of type â…¢ collagen. |
