Objective: To investigate the mechanism of therapeutic effects of Rhein on insulinresistance from aspects of the expression of resistin mRNA, plasma free fatty acid(FFA) andinsulin sensitivity in streptozotocin and diet-induced diabetic rats.Methods: The streptozotocin (STZ) and high fat diet-induced diabetic rats are divided intothree groups by random. They were differently treated with Rhein, pioglitazone and salineby administration by gavage. The expression of resistin mRNA, plasma free fatty acid(FFA),index's antilogarithm of insulin-sensitivity, fasting blood glucose, fasting insulin as well asblood lipid in all groups were detected both before and after administration by gavage for 8weeks.Results: FFA, blood lipid, intake of food and water were significantly decreased (P<0.05) inRhein-treated diabetic obese rats as compared with untreated diabetic obese rats, while theinsulin sensitivity was significantly increased(P<0.01).Conclusion: Rhein-therapy can increase insulin sensitivity, decreased plasma free fatty acid(FFA) and blood lipid.
