[Objective] To establish the specific immunological indexes of sensitized dendritic cells by observing the change of IFN-γ, IL-10 and IL-12 in the peripheral blood of the patients with advanced breast cancer before and after dendritic cell combined with chemotherapy, and observe the clinical curative effect and side effect of the combined chemotherapy in the patients with breast cancer.[Methods] 68 patients with advanced breast cancer were randomly divided into chemotherapy group (control group) and dendritic cells combined with chemotherapy group (observing group), sensitized matured DCs were derived from peripheral blood monocytes of the patients in observing group before chemotherapy which were loaded tumor lysate and cultivated in vitro, in the course of cultivation, the patients in observing group received docetaxel and pirarubicin, sensitized matured DCs were administered after chemotherapy, the patients in control group received the treatment of the same regimen. The concentrations of IFN-γ, IL-10 and IL-12 in blood serum were detected before and after treatment; toxicity, clinical benefit response rate and KPS were observed.[Results] The concentrations of IFN-γ, IL-12 and IL-10 in blood serum and KPS were not significant difference in two groups before treatment; In observing group, the concentrations of IFN-γ, IL-12 in blood serum and KPS were significantly increased, and the concentration of IL-10 was decreased after treatment, in control group, the concentrations of IFN-γ, IL-12 in blood serum and KPS were decreased, and the concentration of IL-10 was increased after chemotherapy, there was statistical difference. No statistical difference was found in clinical benefit response rate between two groups(P>0.05). Compared with control group, bone marrow suppression was mild in obversing group(P<0.05). In observing group, when sensitized matured DCs were administered, fever was a common side effect, serious side effects were not observed.[Conclusions] IFN-γ, IL-10 and IL-12 are the specific immunological indexes of sensitized dendritic cells therapy; The response rate of biochmotherapy is the same as the response rate of chemotherapy; Dendritic cells can decrease side effects of chemotherapy, change immunosuppression, enhance immune function and improve the quality of patients life.
