Objective: To observe the characteristic of subacute methamphetamine hydrochloride in rats. Our target is to observe the changes of mG1uR5 receptors' expression in subacute methamphetamine hydrochloride in the rats. Methods: Forty male Wistar rats were randomly divivided into control group (n=20) and experimental group (n=20) . Animal modle of subacute methamphetamine hydrochloride was induced by intraperitoneally methamphetamine hydrochloride. Rats in the experiment group were intraperitoneal injected with methamphetamine hydrochloride (20mg/kg, 10mg/kg, twice a day at 8:00 and 20:00 respctively for 4 continuous day), and those in the control group were administered with saline of the same volume. MGluR5 receptor were marked used anti-mG1uR5 polyconal antibody by the immunohistochemistry (streptavidin-peroxi-dase, S-P) method. The positive reaction of mG1uR5 neuron in subacute methamphetamine hydrochloride in the rats was qualitatively analyzed by light microscope. The positive reaction of mGluR5 neuron in the subacute methamphetamine hydrochloride in the rats was quantitatively analyzed by image analysis system. The data were analyzed by t-test. Results: 1.Experiment group of locmotor activity, temperature, food-intake, water-intake differered significantly compare to control group. Experiment group of body weight decreased insignificantly compare to control group. 2.Experiment group of the expression of mG1uR5 in the neuron cells of hippocampus, striatum augment differered significantly compare to control group. Experiment group of the expression of mG1uR5 in the neuron cells of cerebral cortex augment was insignificant compared to the control group. Conclusion: 1.The model of subacute methamphetamine hydrochloride in rats can be established successfully by intraperitoneal injection. 2.The mG1uR5 receptors were expressed in the neuron cellular membrane of hippocampus, striatum and cerebral cortex. The expression of mG1uR5 in those regions augment in different degree after intraperitoneal injection methamphetamine hydrochloride found by light microscope. 3.Experiment group of the expression of mG1uR5 in the neuron cells of hippocampus, striatum augment differed significantly compared to the control group. Experiment group of the expression of mG1uR5 in the neuron cells of cerebral cortex augment was insignificant compared to the control group. |
