Objective To investigate the effect of Extract ofGinkgo Biloba (EGB761)on the aged rat myocardial tissue and to studythe related mechanism. Methods 1.The aged rat received gastric infusionof EGB761 for 16 weeks, the control group of aged rat and adult rat wereinfused the same volume saline.2. After they were dissected, we observedthe myocardial histopathological changes under electron microscope.3.we measured the SOD,GSH-Px and MDA in the blood samples usingcolorimetric method.4.AGEs in the myocardial tissue were measured byfluorometric method.5.The mitochondrial DNA (mtDNA) deletion levelin myocardial cell was examined using polymerase chain reaction (PCR).Results 1. The electron microscope showed that myocyte swell, themembrane structure of mitochondria and sarcoplasmic reticulum weredamaged in the control group of aged rat than the administration group.The administration group were similar like the control group of adult rat.2.The contents of the SOD of blood samples in_the aged groups ofadministration of EGB761 and in the control group of nature aged ratwere 211.31±6.83 U/ml vs 138.44±3.80 U/ml (P<0.05). The contents of the GSH-Px of blood samples in_the aged groups of administration ofEGB761 and in the control group of nature aged rat were 1947.11±75.07U/ml vs 1283.80±28.84 U/ml (P<0.05).So the contents of the SOD andGSH-Px of blood samples in_the aging groups of administration of EGB-761 were significantly increased than in the control group of nature agedrat (P<0.05). The contents of the MDA of blood samples in_the agedgroups of administration of EGB761 and in the control group of natureaged rat were 4.24±0.10nmol/ml vs 6.73±0.56nmol/ml (P<0.05). So theMDA level of blood samples is significantly decreased than in thecontrol group of aged rat (P<0.05).3. The contents of the AGEs of themyocardial tissue in_the aged groups of administration of EGB761 and inthe control group of nature aged rat were 24.50±2.26 AU/mgHYP vs33.50±1.29 AU/mgHYP (P<0.05), soAGEs in the myocardial tissueof EGB761 treatment group is significantly decreased than the controlgroup of aged rat. 4. The mitochondrial DNA (mtDNA) deletion level inmyocardial tissue of EGB761 treatment group and in the control group ofnature aged rat were 0.0068±0.0095 vs 0.1805±0.0718(P<0.05), so themitochondrial DNA (mtDNA) deletion level in myocardial tissue ofEGB761 treatment group is significantly decreased than the control groupof aged rat (P<0.05). The control group of adult rat were similar as theadministration group. (P>0.05). Conclusion EGB761 has the anti-agingeffect. The mechanisms may relate with inhibiting nonenzymatic glycation and reducing the free radical.
