Objective: To observe the distribution and the change in the blood-drug concentration of nano-particles iron oxide in mice, which is coated with Glu labeled by 59Fe (nano-59Fe-Fe2O3-Glu). To investigate the toxic reaction in mice.Methods: By coprecipitation to synthesis nano-59Fe-Fe2O3-Glu, the distribution in blood, heart, liver, spleen, kidneys, lungs, brain, eyes and gonad etc are measured, and the character of its pharmacokinetics in vivo is observed, such as the behavior of the absorption, distribution and clearance, accumulation in tissues and the probable target-organ. In the groups of injection of inter-liver, the blood routine test, the biochemical test and the ratio of body weight are determinated, while in the i.v. groups, the micronucleus test and the sperm deformity test are explored.Results: Nano-59Fe-Fe2O3-Glu,the diameter of which is 15nm, the maximum in the liver is 48.9%ID/g and the maximum in the spleen 22.9%ID/g at 1h and 2d respectively after injection of vein of nano-59Fe-Fe2O3-Glu , while in the group of injection of inter-liver the maximum in the liver is 41.3%ID/g,20.8%ID/g,14.6%ID/g and 23.6%ID/g,21.2%ID/g,12.5%ID/g at 8d and 24h in high dose, mid-dose and low dose respectively .There is any radiation in brains, eyes and gonad, which indicates nano-Fe2O3 may diabasis blood brain barrier ,blood testis barrier and so on .The radiation in the blood increases and attains the maximum at 16d both in the group of injection of vein and in the group of injection of inter-liver, and then it decreased ,but it can also be observed at 48d . Compared with the controlled groups, the results of the blood routine test and the biochemical test have no significant difference. There are no effects in the weight of mice after injection of inter-liver, in the micronucleus test and in the sperm deformity test after i.v..Conclusion: The biological distribution of nano-Fe2O3-Glu, which is mainly detained in liver and spleen in vivo, was traced by 59Fe sensitively and accurately, but it has no hemotoxicity, tocytotoxicity and genetic-toxicity .
