Objective:In traditional Chinese medicine, a sort of medicinal herb—solanum lyratum thunb(SLT) has the effect, of clearing away heat and toxic material, promoting blood circulation and removing blood stasis. It is a well-known Chinese traditional medicine used for the treatment of various kinds of cancer, such as liver carcinoma, lung cancer, esophageal carcinoma , gastric cancer and so on. But the action mechanisms of this type of Chinese herbal medicine and its active components are not fully clear. Its apoptosis-inducing and proliferation inhibition effect of no detail was reported so far. Therefore, the purpose of this study is to investigate the apoptosis-inducing and proliferation inhibition effect of solanum lyratum thunb extract and its molecular mechanism on human lung cancer A549 cells, and establish the experimental basis for clinical application. Methods:1. Dried whole herbs of SLT was extracted with boiling distilled water.2. Cultured human lung cancer A549 cells were randomly divided into the control group (the negative control group), SLT-treated groups (10, 20,40mg/ml) and DDP (25mg/L) group (the positive control group).3. The inhibitory rate and IC50 values were evaluated by MTT assay after treated with SLT for 24h, 48h, 72h and 96h.4. After treated human lung cancer A549 cells with SLT for 48h, morphological changes of apoptosis were observed with fluorescance microscope.5. After treated human lung cancer A549 cells with SLT for 48h, induction of cell cycle arrest and apoptotic rate were determined by flow cytometry.
