Objective:1 To investigate the degree and trend of liver injury in severely scalded rats.2 To explore the role of LPS-CD14-TLR-NF-kB pathway after thermal injury.3 To study the effect of PDTC on liver in severely scalded rats. Methods:After they were inflicted with 30% TBSA full-thickness skin scald,the rats were sacrificed and samples were taken.HE stain was used to measure the dgree of liver injury and to count the volume density of hepatocytes,a kind of parameters,by stereological analysis.Immunohistochemistry stain was used to show the expression of NF-kB and CD14 in liver and to determine the numerical density of positive cell. Results:1 Histological changes of liver.In burn group,swelling of hepatocytes was observed at 6 postburn hour,injury increased at 12 postburn hour and necrosis appeared in some lobule,which continued at 24 postburn hour.In burn+PDTC group,slight swelling of hepatocytes at 6 postburn hour,less necrosis at 12 postburn hour,sporadic necrosis in hepatic sinusoid at 24 postburn hour were observed.2 Change of numerical density of CD14 positive cell.The numerical density was 0.1644±0.0037×10-4μm2 in control group,increasing at 6 postburn hour(0.3265 ±0.0179×10-4μm-,p<0.01),peaking at 12 postburn hour (0.3751±0.0171×10-4μm3 P<0.01) ,keeping higher level at 24 postburn hour (0.3624±0.0130×10-4μm3, P<0.01 ) .After treatment with NF-kB inhibitor PDTC the value significantly decreased at 6 12 24 postburn hour ( 0.2750±0.0110×10-4μm3, 0.2945± 0.0020 ×10-4μm3, 0.2906±0.004 ×10-4μm3, P<0.01) compared with corresponding...
