Study About Intervention Of Radix Salviae Miltiorrhizae On Transforming Growth Factor Beta1 In Rats With Acute Lung Injury

Objective: To investigate the effect of Radix Salviae Miltiorrhizae on transforming growth factor beta1(TGF-β₁) in rats with LPS-induced acute lung injury.Methods: LPS(E.Coli O₅₅:B₅)was injected intravenously to form ALI model. Twenty-four healthy Sprague-Dawley rats were randomly divided into 3 groups.Each group has 8 rats .1) ALI group: LPS(5mg/kg) was given by vena caudalis injection.2) control group: LPS was replaced by normal saline.3) Radix Salviae Miltiorrhizae treatment group: LPS(5mg/kg) was given by vena caudalis injection,then an hour later, Radix Salviae Miltiorrhizae (10g/kg) was given by the same way. All the animals were observed about 12 hours after the injection of LPS/normal saline. 2ml anti-coagulation venuous blood was used to investigate hemorheological parameter changes. The animals were sacrificed at the end of experiment by bleeding.The whole lung was taken out from the thorax,and the flood on its surface was sucked in filter paper.The left lung was used for wet and dry weigh ratio(W/D) after it has been baked in the dry up machine about 80℃ for 24 hours. The right upper lung tissue was made into pathological sections which were dyed in hematoxylin-eosin and immunohistochemistry. The pathologicalchanges and TGF-β₁ expression were examined with light microscope in various tissues. The expression of TGF-β₁ was evaluated demiquantitively by H score.Result: 1.observation of lung tissue:From the observation under light microscope,we can see that the interalveolar septum in the lung of ALI group is swollen and wider,having more PMN recruitment.There are also many...
