| PrefaceType 1 diabetes is an organ - specific autoimmune disease mediated by T cell and lung is a target organ of diabetes damaging. Many immunocytes, in which CD4+ T lymphocyte takes major responsibility, participate in the process of diabetes onset. In vivo, Th1 cell secretes IL -2(interleukin 2) , IFN -γ (in-terferon-γ) and TNF-α (tumor necrosis factor - α) etc. On the contrary, Th2 cell secretes IL -4(interleukin 4) and IL - 10 (interleukin 10). We determined Th1 / Th2 immune response status by detection of IFN - γ, IL - 4 and ratio of IFN - γ /IL - 4 in serum and BALF of type 1 diabetes rat model induced by STZ.MethodTwenty male SD rats were equally divided into experimental group (diabetes group) and contrast group (normal group) randomly. Type 1 diabetes rat model was reproduced by administration of streptozotocin to maintain blood glucose between 16.7 and 23.6mmol/L ( blood glucose ≥16.7mmol / L) for one month. After the model was blest, rat was anesthetized and took 5ml blood, which was centrifuged and supernatant was reserved, from aorta abdominalis through open abdomen, and then performed bronchus alveoli lavage and supernatant of BALF was taken. All preserved in - 70℃, refrigerator for wholesale testing. Serum and BALF IFN - γ and IL -4 was detected by ELISA method.Measurement data expressed in mean ±sd ( x|- ± s ) ; t test was adopted for... |
