Objective: explore the role MTA1 may play in the process of tumor progression, invasion and metastasis .Lay the academic basis for further implying RNA interference in anticancer therapy.Methods: construct the recombinant plasmid pGenesil-1/MTA1 carrying shRNA to MTA1, transfect this recombinant plasmid into breast cancer cells and detect the expression of ER and nm23 by immunohistochemistry and RT-PCR.Results:1.The recombinant plasmids suppress MTA1 expression by less than 50% in BCap-37 cell and by more than 70% in MDA-MB-435S cell.2.After siRNAs were transfected in MDA-MB-435s cells, the results showed that ER re-expressed and the expression level of nm23 increased; while there was no effect on the mRNA transcription of ER or nm23 in BCap-37 cells.Conclusion:...
