ObjectiveTo study the differentially expressed gene profile of Heart-Yang deficiency suffering from coronary heart disease, with an emphasis on genes in response to lipid metabolism, energy metabolism and collagen synthesis; and to explore their clinical significance. Methods①Set up the diagnosis criterion of heart-yang deficiency by integrating relevant theories derived from classic literatures, clinic observations and experimental data of our former studies upon deficiency-cold Zheng.②Collected patients under the above diagnosis criterion restrictively, and treated them by Gui-zhi-gan-cao-Tang combined with Bao-yuan-Tang for about 30days.Then we identified the changes before- and after TCM therapy, on index such as the intensity of Heart-Yang deficiency, the level of blood lipid, the blood dynamics and the coefficient of sympathetic nerve.③To study the pattern of the differentially expressed gene profile against Heart-Yang deficiency cases suffering from coronary heart disease, and the pharmaceutical targets at the level of transcriptome, by means of cDNA microarray. Then we mined the clinical significance of these abnormally expressed genes by certain bioinformatic tools, e.g., gene filtration, functional classification, cluster analysis, and exploration of molecular pathway. Results①We set up a diagnosis criterion of Heart-Yang deficiency, which including 25 items of TCM symptoms. Its reliability and validity were tested by standard statistic methods.②24 patients of Heart-Yang deficiency suffering from coronary heart disease were collected in this present study. They displayed significant improvement on almost all the syndromes of Heart-Yang deficiency after TCM treatment. Marked differences were observed on the concentration of blood cholesterin, Apo_A1 and Apo_B, and the coefficient of sympathetic nerve.③ Of the Heart-Yang deficiency suffering from coronary heart disease, 44 pieces of differentially expressed genes were obtained by cDNA microarray approach. (Ratio<0.5 or Ratio>2). And there were 22 genes differentially expressed after TCM treatment(Ratio<0.67 或 Ratio>1 .5).
