Immunological Reaction Of Allogeneic Bone Marrow Stromal Cells Transplantation For The Repair Of Rabbit Radial Bone Defect

Objective To investigate the therapeutic effect and immunological reaction of allogeneic bone marrow stromal cells transplantation for the repair of radial bone defect. Methods A total of 30 4-week-old new Zealand white rabbits were randomly divided into 2 groups of A(fibrin glue~+recombinant bone morphogenetic protein-2~+allogeneic bone marrow stromal cells,each has 15 rabbits.MSCs aspirated from allogeneic rattits' greater troochanter of femur were cultured in vitro and amplified ,then the acquired 3 passages MSCs, rhBMP2 and FG were made into a complex which was implanted in 20 allogenic rabbits 1.5cm segmental bone defects of right radius as experiment group.Fibrin glue only was implanted to the same size defects of right radius as control group.Blood samples were examined for T lymphocyte subsets CD4~+, CD8~+ and CD4~+/CD8~+at the timepoints of the 1st, 2nd, 4th, 6th and 8th postoperatively.the 3 rabbits were euthanized postoperatively at different intervals from 2, 4, 6 and 8 weeks for micropathology and histology examinantion.Results (1) T lymphocyte subsets CD4~+, CD8~+ and CD4~+/CD8~+ at above time points in the experiment group had no significant difference as compared to group; (2) The transplantation of the complex of allogenic bone marrow stromal cells, recombinant bone morphogenetic protein-2 and fibringen causes no obvious immunological rejection reaction; (3) Histological examination after transplantation showed that the bone callus formation of the experimental groups was faster that of the control groups .by the end of...
