| 1. Isolation and Identifiation of the Immunomodulating Peptides Derived Casein Objective To investigate the best condition to prepare immunomodulating peptide from the casein. Set up a method to isolate and identificate immunomodulating peptide from the enzymatic hydrolysis of casein. Methods Casein was hydrolyzed by trysin and chyomtrysin together. The enzymatic hydrolysis was filtrated by ultrafiltrate membrane, and analyzed by SDS-PAGE. Immunomodulating peptide (PGPIPN) was identificated in the enzymatic hydrolysis by RP-HPLC in external standard method and internal standard method, use PGPIPN as the standard. Then the enzymatic hydrolysis of the casein was chromatographed by Sephadex G-15, each pink of which was determined if there is PGPIPN by RP-HPLC. Results PGPIPN could be preparated from casein by enzymatic hydrolysis of trysin and chyomtrysin together, which yield was about 5%. The production of enzymatic hydrolysis was chromatographed by Sephedax G-15, PGPIPN emerged in the 5th pink of fractions. Conclusion Casein enzymatic hydrolyze by the trypsin and chrometrypin can form the immunomodulating peptide, and the immunomodulating peptide can be isolated by Sephadex G-15.2.
